辣椒脱落酸合成相关酶基因的鉴定与表达分析

摘　　要：类胡萝卜素裂解双加氧酶基因家族的9-顺-环氧类胡萝卜素双加氧酶(NCED)是脱落酸合成的关键酶类。利用生物信息学方法,鉴定辣椒脱落酸合成相关酶家族基因,并对基因结构、进化、功能和时空表达进行分析。研究发现,辣椒基因组中包含3个NCED基因和6个CCD基因,NCED基因不含内含子,CCD基因含有多个内含子;归为5个聚类组,与同为双子叶植物的拟南芥基因亲缘关系较近;CaNCED1具有与VP14类似的三维结构和催化基团;基因启动子区发现多个激素响应元件、环境胁迫响应元件;CaNCED1、CaCCD1a和CaCCD1b在辣椒各器官和果实成熟的各阶段均显示较高表达。该研究为辣椒ABA合成相关酶基因的功能分析和遗传育种应用提供了科学依据。9-cis-epoxy carotenoid dioxygenase (NCED), a member of carotenoid cleavage dioxygenases (CCD) superfamily, is a key enzyme in the synthesis of abscisic acid. The gene structure, evolution, function and spatiotemporal expression of CCD family genes in pepper were analyzed by bioinformatics. The study found that the pepper genome contained three NCED genes and six CCD genes, and NCED genes contained no intron, CCD genes contained multiple introns. Pepper CCD family was classified into 5 clusters, and kept a more recent relationship with dicot Arabidopsis. CaNCED1 had a similar three-dimensional structure and catalytic group with VP14. Multiple hormone responsive elements and environmental stress response elements were found in the gene promoter region. CaNCED1, CaCCD1a, and CaCCD1b showed high expression in different pepper organs and different fruit ripening stages. This study provided theoretical support for functional analysis and genetic breeding application of pepper ABA synthesis key enzyme genes.
