青蒿琥酯治疗类风湿关节炎的机制研究进展

摘　　要：类风湿关节炎(RA)是一种以关节破坏为特征的慢性炎症性自身免疫病,目前缺乏高效低毒的药物.青蒿琥酯是抗疟的一线药物,新近发现其对自身免疫病尤其是RA具有治疗效果.该文综述了青蒿琥酯治疗RA的机制研究,证实青蒿琥酯不仅能调节T、B淋巴细胞的免疫功能,抑制自身抗体形成,还可直接作用于成纤维样滑膜细胞和破骨细胞,抑制滑膜炎症、新生血管形成及破骨细胞分化活化,抑制RA关节炎症和骨破坏,有望进一步在临床应用.Rheumatoid arthritis(RA)is a chronic inflammatory autoimmune disease characterized by the destruction of joints.Currently,an efficacious and safe drug for RA is still lacking.Artesunate,a firstline drug for malaria,was recently found to have potential in treating autoimmune diseases,especially RA.In this article,the pharmacological mechanism of artesunate in the treatment of RA was summarized.The findings demonstrated that artesunate could not only regulate the immune function of T cells and B cells and suppress the formation of autoantibodies,but also directly affect the fibroblast-like synoviocytes and osteoclasts,inhibit the synovitis,neovascularization and osteoclastogenesis,and then suppress joint inflammation and bone destruction of RA.Therefore,artesunate might be a potential drug for RA in clinical practice.
