基于流式细胞技术分析碲化镉量子点对小鼠生精过程的影响

摘  要:为探讨碲化镉对雄性小鼠生殖影响机理,将27只雄性小鼠随机分为3组,采用静脉注射的方法分别注射0、0.2和2nmol的碲化镉,在处理后第1天、3天和7天,处死小鼠,摘取睾丸后制作细胞悬液,采用PI染色法及流式细胞分析技术,检测碲化镉量子点对小鼠睾丸内各级生精细胞组成的影响。实验结果表明,碲化镉显著影响了睾丸内生精细胞的构成,处理组的睾丸内单倍体精子细胞和精子下降,四倍体的初级精母细胞升高,精子发生受到影响。本实验为深入阐明碲化镉对雄性小鼠生殖毒性的机制提供试验依据,并为量子点的生物安全评价和生物医学应用提供指导。To investigate the mechanism of cadmium telluride on the reproduction of male mice, 27 male mice were randomly divided into 3 groups. Two experimental groups were injected intravenously by 0.2 nmol and 2 nmol of cadmium telluride (CdTe). Mice were sacrificed on day 1, 3 and 7 after treatment. Testicular cell suspensions were made for PI staining and flow cytometry analysis. The effect of CdTe on spermatogenic cells in spermatozoa of mice was uncovered. The results showed that CdTe significantly affected the composition of spermatogenic cells in the testis. In the experimental group, the haploid sperm cells and sperm decreased, and the tetraploid primary spermatocytes were high, which indicted spermatogenesis was affected. This experiment is helpful for further clarification of the mechanism of CdTe on reproductive toxicity in male mice. Meanwhile, it will guide the biosafety evaluation and biomedical applications of quantum dots.
