摘 要:目的研究慢性宫内缺氧对子代主动脉microRNA (miRNA)表达谱的影响.方法将16只金黄地鼠孕鼠随机分为宫内缺氧组(CIH)和正常对照组(NC),取4周龄子代主动脉进行数字化基因表达谱测序,并进行qRT-PCR验证.结果与NC组相比,CIH组共有9个miRNA上调,10个miRNA下调,靶基因涉及多种代谢酶及炎症因子;KEGG Pathway分析显示差异miRNA在多个炎症及氧化应激相关信号通路富集明显.结论 CIH可诱导子代主动脉miRNA表达谱发生改变,并可能通过差异miRNA参与其子代成年期心血管疾病的发生发展.Objective To research the effect of chronic intrauterine hypoxia ( CIH) on offspring aorta microRNA ( miRNA) expression profile. Methods Sixteen pregnant golden hamsters were randomly divided into CIH group and normal control ( NC) group. Differentially expressed miRNA of aorta in 4-week-old offsprings were examined and identified by RT-PCT. Results Compared with NC group, 9 miRNAs were upregulated and 10 miRNAs were down-regulated in CIH group, the target genes involved multiple metabolic enzymes and inflammatory factors. KEGG Pathway analysis showed that differential miRNAs were significantly enriched in multiple inflammatory pathways and oxidative stress signaling pathways. Conclusion CIH leads to the changes of miRNA expression profile in aorta of offsprings, which may be the possible mechanism of CIH related cardiovascular diseases in adulthood of offsprings .
