摘  要:【目的】采用线粒体COI基因序列分析武汉地区常见麦蚜蚜种的遗传组成,探讨基于COI基因序列作为常见麦蚜蚜种分子鉴定的可行性,为田间麦蚜的有效识别和综合治理提供科学依据。【方法】采集武汉市田间和实验室培养的3种麦蚜(禾谷缢管蚜、麦长管蚜和麦二叉蚜),扩增其COI基因后进行测序,并进行遗传距离检测及聚类分析。【结果】3种麦蚜COI基因片段核苷酸组成中碱基C和G的总含量低于碱基A和T的总含量,其中含量最低的碱基为G,具有明显的碱基偏倚性;不同麦蚜的种内遗传距离均小于0.010,种间遗传距离约是种内遗传距离的10倍;聚类分析结果表明,蚜科的所有物种聚为一支,之后又形成3个分支,分属于蚜科的3种麦蚜。【结论】3种麦蚜的分子鉴定与形态学分类结果一致,表明采用COI基因序列可对武汉地区常见麦蚜蚜种进行有效鉴定。【Objective】This experiment used mitochondrial COI sequences to analyze the genetic composition of common wheat aphids species in Wuhan,investigated the feasibility of COI gene sequence as molecular identification for common wheat aphid species,and provided reference for effective identification and integrated management of wheat aphids in the field.【Method】Samples of three different wheat aphids species[Rhopalosiphum padi(Linnaeus),Macrosiphum avenae(Fabricius)and Schizaphis graminum]in the field in Wuhan and cultivated in the laboratory were collected. The COI gene were amplified and sequenced. Genetic distance and the clustering were also analyzed.【Result】In the nucleotide of COI gene fragment of the three wheat aphids,the total content of base C and base G was lower than the total content of base A and base T. The content of base G was the lowest and showed obvious base bias. All intraspecific genetic distances of these aphid species were lower than 0.010,and interspecific genetic distance was ten times more than intraspecific genetic distance of these aphid species. The analysis of clustering showed that all species of Aphidinae were clustered one clade,and then it formed three small clades which were individually clustered by different wheat aphids.【Conclusion 】The molecular identification of these wheat aphids in the field is consistent with the morphological classification. The results suggests that COI gene sequence can identify effectively the common wheat aphids inWuhan.
