摘 要:目的:探讨黄秋葵醇提物对链脲佐菌素诱导的糖尿病小鼠胰腺组织中氧化应激的影响。方法:给予昆明种小鼠尾静脉注射120 mg/kg链脲佐菌素,以空腹血糖(FBG)≥11.1 mmol/L视为糖尿病模型小鼠,随后将糖尿病小鼠随机分为模型组、二甲双胍组及黄秋葵醇提物低、中、高剂量(0.2 g/kg、0.4 g/kg、0.8 g/kg)组,并另取健康昆明种小鼠作为正常组。分组后的动物灌胃给予相应药物和生理盐水14 d。检测小鼠血清和胰腺组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性和丙二醛(MDA)含量;采用酶联免疫吸附试验(ELISA)法检测小鼠胰腺组织中氧化氢酶(CAT)、活性氧簇(ROS)的含量;苏木精-伊红(HE)染色观察小鼠胰腺组织病理形态学变化。结果:与模型组比较,二甲双胍组和黄秋葵醇提物组FBG均有不同程度的下降(均P<0.05);黄秋葵醇提物组血清和胰腺组织中SOD和GSH-Px活性明显升高,MDA含量下降,胰腺组织中CAT活性升高,ROS含量下降(均P<0.05);胰腺HE染色结果显示,黄秋葵醇提物组胰腺组织病理变化明显改善。结论:黄秋葵醇提物具有降糖作用,可提高糖尿病小鼠血清和胰腺组织中SOD、GSH-Px、CAT抗氧化酶活性,降低MDA和ROS含量,从而缓解氧化应激对糖尿病小鼠胰腺组织的损伤作用。Objective: To investigate the effect of ethonal extracts of Abelmoschus esculentus Moench on oxidative stress of pancreatic tissues in diabetes mellitus mice. Methods: Kunming mice were injected with 120 mg/kg streptozotocin through tail vein.Mice with fasting blood glucose (FBG)≥11.1 mmol/L were considered as diabetes and were randomly divided into model control group,metformin group,and ethonal extracts of Abelmoschus esculentus Moench groups (0.2,0.4,0.8 mg/kg).Healthy Kunming mice were considered as normal control group.Mice were given drugs and saline for 14 days.The superoxide dismutase (SOD),glutathione peroxidase (GSH-Px) activities and malondialdehyde (MDA) levels in serum and pancreatic tissues were determined.ELISA method was used to detect the hydrogenase (CAT) and reactive oxygen species (ROS) contents in pancreatic tissues.Pathomorphological changes were observed by HE staining. Results: Compared with model group,the FBG was decreased after mice treated with metformin and ethonal extracts of Abelmoschus esculentus Moench .The SOD and GSH-Px activities in serum and pancreatic tissues were enhanced,while the MDA content was decreased ( P <0.05).The activity of CAT increased and the content of ROS decreased in pancreatic tissues ( P <0.05).HE results showed that the pathological changes in ethonal extracts of Abelmoschus esculentus Moench groups were alleviated compared with model group. Conclusion: Ethonal extracts of Abelmoschus esculentus Moench has an anti-diabetic effect.It could mitigate pancreatic damage induced by MDA and ROS through enhancing the activities of SOD,GSH-Px and CAT,resulting in oxidative stress reduction on diabetic mice pancreas.
