摘  要:目的分析右美托咪定对心肌缺血再灌注(MIR)大鼠模型心肌梗死面积、心肌细胞凋亡及凋亡蛋白表达的影响。方法取30只SD大鼠,随机分为对照组(仅穿线而不结扎)、MIR组(建立MIR模型)、干预组(建立MIR模型前给予右美托咪定预处理),三组各10只。比较干预组与MIR组大鼠心肌梗死面积、危险区面积和心肌细胞凋亡率的差异,并比较三组大鼠心肌组织中B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)及补体3蛋白表达水平的差异。结果相比MIR组,干预组大鼠心肌梗死面积和危险区面积明显缩小,心肌细胞凋亡率显著降低(P <0. 01)。相比对照组,MIR组和干预组大鼠Bcl-2和Bax蛋白表达水平均显著升高(P <0. 01);相比MIR组,干预组大鼠Bcl-2蛋白表达水平显著升高,而Bax蛋白表达水平显著降低(P <0. 01)。相比对照组,MIR组和干预组大鼠补体3蛋白表达水平均显著升高(P <0. 01);相比MIR组,干预组大鼠补体3蛋白表达水平显著降低(P <0. 01)。结论通过右美托咪定预处理可提高MIR大鼠模型心肌Bcl-2蛋白表达,降低Bax和补体3蛋白表达,减少心肌梗死面积,缓解大鼠心肌损伤,减少心肌细胞凋亡。Objective To investigate the effects of dexmedetomidine on myocardial infarction size, apoptosis and apoptotic protein expression in rats with myocardial ischemia-reperfusion (MIR). Methods Thirty SD rats were randomly divided into three groups including control group (only threading without ligation), MIR group (establishment of MIR model), intervention group (pre-treatment with dexmedetomidine before establishment of MIR model), 10 rats in each group. The differences of myocardial infarction area, risk area and apoptotic rate of myocardial cells between intervention group and MIR group were compared. The expressions of B-cell lymphoma-2 (Bcl-2), Bcl-2 associated X protein (Bax) and complement 3 protein in three groups were also compared. Results Compared with MIR group, the area of myocardial infarction and risk area in intervention group were significantly reduced, and the apoptotic rate of myocardial cells was significantly reduced ( P <0.01). Compared with the control group, the expression of Bcl-2 and Bax protein in MIR group and intervention group increased significantly ( P <0.01);compared with MIR group, the expression of Bcl-2 protein in intervention group increased significantly, while the expression of Bax protein decreased significantly ( P <0.01). Compared with the control group, the expression level of complement 3 protein in MIR group and intervention group was significantly increased ( P <0.01);compared with MIR group, the expression level of complement 3 protein in intervention group was significantly decreased ( P <0.01). Compared with MIR group, the area of myocardial infarction and risk area in intervention group were significantly reduced, and the apoptotic rate of myocardial cells was significantly reduced ( P <0.01).
