苏铁叶总黄酮联合顺铂对Lewis肺癌小鼠的抑瘤作用

摘　　要：目的研究苏铁叶总黄酮联合顺铂对Lewis肺癌小鼠的抑瘤作用。方法将50只C57BL/6小鼠按照体重随机分为正常组、模型组、顺铂组、中药组及联合组,每组10只。除正常组外,其余小鼠均于右下肢腋窝处注射Lewis细胞悬液0.2mL构建Lewis肺癌移植小鼠模型,接种7d后,顺铂组每周腹腔注射顺铂0.4mL+每日灌胃0.9%NaCl0.2mL;中药组每日灌胃苏铁叶总黄酮0.4mg·kg-1·d-1;联合组每日灌胃苏铁叶总黄酮0.4mg·kg-1·d-1+每周腹腔注射顺铂0.4mL,正常组正常饲喂,不给予药物干预,模型组每日灌胃0.9%NaCl+每周腹腔注射0.9%NaCl,均连续干预21d。检测各组小鼠肿瘤生长及转移情况;检测各组小鼠胸腺指数(TI)、脾指数(SI),以噻唑蓝(MTT)法检测各组小鼠脾T、B淋巴细胞增殖水平;以免疫组化法检测小鼠肺肿瘤组织血管内皮生长因子(VEGF)蛋白表达。结果顺铂组、中药组和联合组小鼠抑瘤率(TIR)分别为(48.33±5.16)%,(52.12±5.32)%,(64.63±2.34)%,肺转移灶抑制率分别为(57.34±2.65)%,(52.27±1.59)%,(76.92±1.49)%,小鼠肺肿瘤组织VEGF蛋白免疫组化染色评分分别为(6.24±0.51),(6.31±0.51),(5.56±0.25)分。药物干预结束后,与模型组比较,其他药物干预组小鼠的肿瘤体积及重量均缩小/降低,与顺铂组比较,联合组小鼠的肿瘤体积、重量缩小/降低,TIR升高(P<0.05)。与模型组比较,其他药物干预组小鼠肺转移率降低,肺转移病灶数量减少(P<0.05),与顺铂组比较,联合组小鼠转移病灶数量减少,肺转移灶抑制率升高(P<0.05)。与模型组比较,顺铂组TI、SI、脾淋巴细胞增殖活性降低(P<0.05);与顺铂组比较,中药组和联合组TI、SI、脾淋巴细胞增殖活性升高(P<0.05)。与模型组比较,顺铂组、中药组和联合组VEGF蛋白免疫组化染色评分降低(P<0.05);与顺铂组比较,联合组VEGF蛋白免疫组化染色评分降低(P<0.05)。结论苏铁叶总黄酮联合顺铂可抑制Lewis肺癌小鼠肿瘤生长及转�Objective To investigate the antitumor effect of total flavonoids of Cycas leaves and cisplatin on Lewis lung cancer mice. Method Fifty C57 BL/6 mice were randomly divided into normal group, model group, cisplatin group, traditional Chinese medicine(TCM) group and combination group according to body weight, each group had 10 mice, except the normal group, the other mice were injected Lewis cell suspension 0.2 mL into the axillary of right lower limb to establish Lewis lung cancer transplantation mice model, after 7 d of inoculation,cisplatin group was intraperitoneally injected with cisplatin 0. 4 m L per week + 0. 9% NaCl 0. 2 m L orally per day;TCM group was given total flavonoids 0. 4 mg·kg-1 orally per day;combination group was given total flavonoids 0. 4 mg·kg-1 orally per day + intraperitoneally injected with cisplatin 0. 4 m L per week;normal group was fed normally without drug intervention;model group was given 0. 9% NaCl orally per day + intraperitoneally injected 0. 9% NaCl per week,all were intervented for 21 d. Tumor growth and metastasis were detected in each group,thymus index( TI),spleen index( SI) and proliferation of spleen T and B lymphocytes were detected by MTT,and expression of vascular endothelial growth factor( VEGF) protein in lung tumor tissue was detected by immunohistochemistry. Results The inhibitory rates of( TIR) in cisplatin group, TCM group and combination group were( 48. 33 ± 5. 16)%,( 52. 12 ±5. 32)%,( 64. 63 ± 2. 34)%,and lung metastasis were( 57. 34 ± 2. 65)%,( 52. 27 ±1. 59)%,( 76. 92 ±1. 49)%,the immunohistochemical staining score of VEGF protein were( 6. 24 ± 0. 51),( 6. 31 ± 0. 51),( 5. 56 ±0. 25) score. Compared with model group,the tumor volume and weight of the mice in other drug intervention group reduced/decreased,and compared with cisplatin group,the tumor volume and weight of the mice in combination group reduced/decreased,and the TIR increased( P < 0. 05).
