摘  要:目的建立HPLC法测定ATP竞争性mTOR抑制剂INK128的含量。方法采用Lichrospher CN(250mm×4.6mm,5μm)色谱柱;以乙腈为流动相A,以0.01mol·mL^-1 磷酸二氢铵为流动相B,进行梯度洗脱(0~40min,5% A→15% A;40~50min,15% A→5% A),流速1.0mL·min^-1 ,检测波长247nm;柱温30℃,进样量20μL。结果 INK128在50.16~802.56μg·mL^-1 范围内与色谱峰面积呈良好线性关系;精密度RSD为0.29%;平均回收率(n=9)为99.6%。结论本方法准确可靠,重复性好,可用于INK128的质量控制。OBJECTIVE To establish a HPLC method to determine ATP-competitive mTOR inhibitor INK128. METHODS The analysis was performed on a Lichrospher CN column (250mm×4.6mm,5μm)with gradient mobile phase of acetonitrile-0.01mol·mL^-1 ammonium dihydrogen phosphate solution at a flow rate of 1.0mL·min^-1 .The detection wavelength was set at 247nm,and the column temperature was 30℃,the injection volume was 20μL.RESUITS The linear range of INK128 was 50.16~802.56μg·mL^-1 ,the precision RSD was 0.29%,and the average recoveries(n=9) was 99.6%. CONCLUSION The method is accurate and good repeatability,can be used for quality control of INK128.
