摘 要:目的建立不同产地蛇足石杉中石杉碱甲含量的测定方法。方法蛇足石杉经乙醇超声提取后,用RP-HPLC测定。采用Waters sunfire C 18 (4.6×250mm,5μm)色谱柱,流动相为甲醇-0.08moL·L ^-1 醋酸铵缓冲液(pH=6.0)(35∶ 65);流速为0.8mL·min^-1;检测波长为310nm。结果石杉碱甲在0.204~25.50μg·mL^-1 与峰面积呈良好的线性关系(r=0.9999);石杉碱甲的平均回收率为100.16%(RSD=1.32%,n=9)。结论该方法快速、简便,结果准确、可靠,可用于蛇足石杉中有效成分石杉碱甲的含量测定。OBJECTIVE To develop a HPLC method for determination of Huperzine A in Huperziaserrate (Thunb.) Trev. METHODS Huperziaserrate (Thunb.) Trev was extract after ultrasonic extraction of ethanol.The analysis was performed on a Waters sunfire C 18 (4.6×250mm,5μm) column with the mixture of methanol-0.08moL·L^-1 ammonium acetate buffer (pH 6.0)(35∶ 65)as mobile phase,and the flow rate was 0.8mL·min -1;The detection wavelength was 310nm;The column temperature was 30℃. RESULTS The calibration curves showed good linearity in the range of 0.204~25.50μg·mL ^-1 (r=0.9999).The average recoveries was 100.16%(RSD=1.32%,n=9). CONCLUSION This method was simple,accurate and reliable to determine the Huperzine A in Huperzia serrate (Thunb.) Trev for the quality control.
