摘 要:目的建立高效液相色谱法测定山绿茶降压胶囊中绿原酸的含量、紫外-可见分光广度法测定其中绿原酸的溶出度,为山绿茶的质量控制提供依据。方法 HPLC:采用十八烷基硅烷键合硅胶为填充剂;乙腈-0.4%磷酸(15∶ 85)为流动相,检测波长327nm,进样量10μL。UV-Vis:经溶出方法考察,最终确定以0.1mol·L -1 盐酸溶液为溶出介质,转速为100r·min ^-1 ,用紫外测定。结果 HPLC:绿原酸的浓度在2.4~12.0μg·mL^-1 范围内线性关系良好,以绿原酸质量浓度(μg·mL ^-1 )为横坐标,绿原酸峰面积为纵坐标,进行线性回归,得回归方程Y=43.339X-6.362,r=0.9995(n=5)。精密度RSD%为2.21%、重复性RSD%为2.56%、稳定性RSD%为2.23%、回收率RSD%为0.57%,平均回收率为96.38%。UV-Vis:山绿茶降压胶囊的绿原酸溶出度符合《中国药典》要求。结论该法快速、准确,操作简便,可用于该胶囊的质量控制。OBJECTIVE To establish a HPLC method for the determination of chlorogenic acid in the antihypertensive capsules of Camellia green tea and to determine the dissolution of chlorogenic acid by UV-vis spectrophotometry,and to provide a basis for quality control of green tea. METHODS The 18 alkyl silane bonded silica gel was used as filler,acetonitrile-0.4% phosphoric acid (15∶ 85) was the mobile phase,the wavelength 327nm was detected and the put 10μL was measured.UV-vis:After the dissolution of the method,the final determination of 0.1mol·L^-1 hydrochloride solution for the dissolution of the medium,rotational speed of 100r·min^-1 ,by ultraviolet determination. RESULTS The concentrations of chlorogenic acid in 2.4~12.0μg·mL ^-1 range has good linear relations,with chlorogenic acid mass concentration (μg·mL ^-1 ) as horizontal axis,chlorogenic acid peak area as ordinate,linear regression,regression equation y=43.339x-6.362,r=0.9995(n=5).The precision RSD was 2.21%,the repeatability RSD was 2.56%,the stability RSD was 2.23%,the recovery RSD was 0.57%,and the average recovery rate was 96.38%.UV-vis:Camellia Green tea step-down capsules of chlorogenic acid dissolution in line with the “Chinese Pharmacopoeia” requirements. CONCLUSION The method is quick,accurate and easy to operate,and can be used to control the quality of the capsule.
