积芎解毒方对糖尿病肾病大鼠肾组织NF-кB、AngⅡ表达的影响

摘　　要：目的:观察积芎解毒方对糖尿病肾病大鼠肾组织NF-кB、AngⅡ表达的影响。方法:将雄性SD大鼠随机分为正常对照组、模型对照组、中药高剂量组、中药中剂量组、中药低剂量组、福辛普利组。除正常对照组外采用STZ腹腔注射制作糖尿病肾病大鼠模型,8周后检测大鼠24 h UTP,Real time PCR及Western Blot法检测大鼠肾组织NF-кB、AngⅡmRNA和蛋白的表达情况。结果:与正常对照组比较,模型对照组大鼠肾组织NF-кB、AngⅡmRNA和蛋白的表达显著升高;与模型对照组比较,各治疗组24 h UTP明显下降,NF-кB、AngⅡ和蛋白表达均明显降低。结论:积芎解毒方高剂量对糖尿病肾病大鼠的肾保护作用可能与通过降低炎症反应从而延缓肾功能发展有关。Objective: To observe the effects of Ji-xiong detoxifcation prescription on NF-кB,AngⅡ expression of diabetic nephropathy rats.Methods: SD rats were randomly selected 10 for normal group,the remaining 50 were dealed with the method of intraperitoneal injection of streptozotocin(STZ) to copy the diabetic nephropathy rat model,then randomly divided into Model group,Ji-xiong detoxifcation prescription(high dose,middle dose, low dose) group and fosinopril group,rats in the 8 th weekend testing 24 hours UTP;Real time PCR was used to detect the expression of NF-кB and AngⅡ mRNA,and Western blot was used to detect the expression of NF-кB and AngⅡ protein. Results: Compared with normal group,NF-кB, AngⅡ mRNA and protein expression quantity of model group were significantly increased;Compared with model group, UTP in the treatment groups lower,NF-кB,AngⅡ mRNA and protein expression quantity obviously decrecse. Conclusion: The renal protection of Ji-xiong detoxifcation prescription on diabetic nephropathy rats may be related to lower inflammatory reaction.
