摘  要:为筛选出杉木苗组织培养适宜的外植体类型及其消毒方法,对外植体灭菌时间、外植体类型和取材时间等进行研究。结果表明:采用0.1%的升汞消毒10min最佳,萌芽率达51.11%;去针叶茎段和茎尖的萌芽率分别为40.00%和8.33%,二者差异显著。去针叶茎段为较适宜的外植体。结果还表明:春季取材优于秋季取材,且苗龄小、木质化低,有利于无菌体系建立。最佳无菌体系建立的方法:在春季选择1a苗龄的杉木苗,去除茎尖和针叶的茎段,先用75%的酒精浸泡15~30s,再用0.1%的升汞消毒10min后接入培养基。该方法感染率和死亡率低,萌芽率最高,依次为14.44%、12.22%和73.33%。In order to select suitable types of explants from Chinese fir seedlings and their disinfection methods for tissue culture, the time of explant disinfection, types of explants and the season of selecting explants were investigated. The results showed that the explants had a germination rate 51.11% after disinfected with 0.1% HgCl2 for 10 min. The germination rates of stem sections with no leaves, and shoot tips were 40.00% and 8.33%, respectively. The stem sections with no leaves were the best explants. The results also showed that the explants collected in spring were better than those collected in autumn in that their seedlings were young and less lignified, which is favorable for establishment of contamination-free system. The optimum contamination-free system was established as follows: select 1 year old Chinese fir seedlings in spring, remove shoot tip and needle leaves of the stems, soak the stems for 15-30 s with 75% alcohol, and disinfect the stems with 10% mercury chloride for 10 minutes. With this optimum method the contamination rate and mortality rate were low, 14.44% and 12.22% respectively, and the germination rate was the highest, up to 73.33%.
