地黄配方颗粒HPLC特征图谱研究及其毛蕊花糖苷含量测定

摘　　要：[目的]建立地黄配方颗粒的HPLC特征图谱,并测定其毛蕊花糖苷的含量。[方法]以流动相乙腈-0.1%冰醋酸水溶液,梯度洗脱,柱温25℃;体积流量1.0mL/min;检测波长334nm;进样量10μL。[结果]建立了地黄配方颗粒的HPLC特征图谱,确定8个特征峰;毛蕊花糖苷在4.89~195.68μg/mL(R2=0.9996)与色谱峰面积呈良好的线性关系;其平均加样回收率为101.57%,RSD为1.29%。[结论]该方法简单、准确、重复性好,为地黄配方颗粒的工艺研究及质量控制提供了参考依据。[Objective]The research aimed to establish the HPLC specific chromatogram of rehmannia formula granule and to determine acteoside content.[Method] For characteristic chromatograms,chromatographic experiments were performed with the mobile phase of 0.1% acetic acid-acetonitril in gradient elutione and the column temperature was set at 25 ℃. The detection wave length was 334 nm and the flow rate was 1.0 mL/min.[Result] The HPLC specific chromatogram of rehmannia formula granule showed 8 characteristic peaks,and the linear range of acteoside was 4.89-195.68 μg/mL( R 2=0.999 6), with average recovery at 101.57%(RSD=1.29%).[Conclusion] This method is simple, accurate and good reproducibility, and can provide a reference for the technical study and quality control of rehmannia formula granule.
