摘 要:本试验旨在研究低磷浓度下原代培养肉鸡鸡胚十二指肠上皮细胞对磷吸收及相关磷转运载体的表达。试验采用单因子完全随机设计,设0(对照组)、0.5、1.0、2.0和4.0 mmol/L 5个不同磷浓度组,每组6个重复,孵育时间为87 min。结果表明:不同磷浓度对十二指肠上皮细胞中磷吸收率有显著影响(P<0.05),磷吸收率随着磷浓度的增加呈线性或二次曲线增加(P<0.05)。与对照组相比,4.0 mmol/L组的十二指肠上皮细胞中Ⅱb型钠磷协同转运载体(NaPi-Ⅱb)和无机磷转运载体1(PiT-1)的mRNA相对表达量显著降低(P<0.05),而4.0 mmol/L组的十二指肠上皮细胞中无机磷转运载体2(PiT-2)的mRNA相对表达量无显著变化(P>0.05)。不同磷浓度对十二指肠上皮细胞中NaPi-Ⅱb、PiT-1和PiT-2蛋白表达无显著影响(P>0.05)。由此可见,随着孵育液磷浓度的增加,细胞磷吸收率增加;当孵育液中磷浓度为4.0 mmol/L时,原代培养肉鸡鸡胚十二指肠上皮细胞可通过下调NaPi-Ⅱb和PiT-1的mRNA表达来调节磷的吸收。The aim of the study was to investigate the phosphorus absorption and related transporter expression under low phosphorus concentrations in primarily cultured duodenal epithelial cells of broiler embryos. A singlefactor completely randomized design was used in this experiment, the cells were randomly assigned to 5 groups with 6 replicates per group, and then treated with 0 (control group), 0.5, 1.0, 2.0 and 4.0 mmol/L phosphorus for 87 min, respectively. The results showed that different phosphorus concentrations had significant effect on phosphorus absorption percentage in duodenal epithelial cells (P<0.05), and the phosphorus absorption percentage was linear or quadratic increased with phosphorus concentration increased (P<0.05). Compared with the control group, the mRNA relative expressions of type Ⅱb sodium-phosphate cotransporter (NaPi-Ⅱb) and inorganic phosphate transporter 1 (PiT-1) in duodenal epithelial cells of 4.0 mmol/L group were significantly decreased (P<0.05), but the mRNA relative expression of inorganic phosphate transporter 2 (PiT-2) in duodenal epithelial cells of 4.0 mmol/L group had no significant change (P>0.05). Different phosphorus concentrations had no significant effects on the protein expressions of NaPi-Ⅱb, PiT-1 and PiT-2 in duodenal epithelial cells (P>0.05). In conclusion, with the incubation solution phosphorus concentration increased, the cell phosphorus absorption percentage increase;when the incubation solution phosphorus concentration is 4.0 mmol/L, the primarily cultured duodenal epithelial cells of broiler embryos can regulate the phosphorus absorption by reducing the mRNA expressions of NaPi-Ⅱb and PiT-1.
