摘 要:【目的】用醋酸泼尼松灌胃处理小鼠建立免疫抑制动物模型,以贯筋藤为材料,考察贯筋藤提取物对正常小鼠及免疫低下小鼠免疫功能的影响.【方法】采用乙醇辅助超声萃取的方法制备贯筋藤粗提物,将小鼠按体质量随机分为空白组、贯筋藤醇提物组、贯筋藤料渣醇提物组、模型组、贯筋藤醇提物+醋酸泼尼松组、贯筋藤料渣醇提物+醋酸泼尼松组6组.空白组灌胃给予同样体积的羧甲基纤维素钠溶液,模型组除给予一定体积的羧甲基纤维素钠溶液外还给予一定量的醋酸泼尼松,连续灌胃给药2周,通过小鼠碳粒廓清试验,测定小鼠体质量、廓清指数、吞噬指数、免疫器官(胸腺和脾脏)等指标.【结果】与空白组相比,贯筋藤醇提物处理均能明显提高正常小鼠脾脏指数、胸腺指数、巨噬细胞吞噬指数和廓清指数,且有统计学差异(P<0.05);与模型组相比,贯筋藤提取物对免疫抑制小鼠免疫器官(胸腺和脾脏)指数、巨噬细胞的廓清指数和吞噬指数均有显著增强作用,且有统计学差异(P<0.05或P<0.01);模型组与空白组比较,小鼠体质量显著下降且有统计学差异(P<0.01),廓清指数明显降低且有统计学差异(P<0.05),吞噬指数有所降低但无统计学差异(P>0.05).【结论】贯筋藤醇提物对小鼠免疫功能有一定的促进作用.【Objective】 In order to establish immunodepressive animal model,the mice were oral administered with prednisone acetate continuously for two weeks.Immunoregulation effecst of ethanol extract of Dregea sinensis on the immunodepressive and normal mice were tested.【Method】 The crude extract was prepared by alcohol and ultrasonic extraction method.The SPF Kunming mice were randomly divided into six groups, including prednisone acetate group(also called immunodepressive model group), control group,ethanol extrat of Dregea sinensis Hemsl(EEDSH)group,ethanol extrat of residue of Dregea sinensis Hemsl(EERDSH)group, EEDSH+prednisone acetate group, EERDSH+prednisone acetate group. The control group was given sodium carboxymethyl cellulose(CMC-Na) solution by gavage,and the immunodepressive model groups were given CMC-Na and prednisone acetate by gavage.The EEDSH and the EERDSH were suspended in the CMC-Na solution and given to mice by gavage.Each group was oral administered corresponding drugs two weeks.The body weight,immune organs indexes,macrophage swallow function and carbon clearance capacity indexes were determined.【Result】 Compared with the control group,the EEDSH group significantly enhanced spleen index,thynus index,macrophage phagocytosis index and expurgation index(P<0.05). Compared with the model group,EEDSH increased the immune organ indexes,expurgation index and phagocytic index(P<0.01,P<0.05).Comparing with the control,the weight and expurgation index of the model group significantly decreased.【Conclusion】 The alcohol extract of the Dregea Sinensis Hemsl has some effect on the immune function of mice.
