摘　　要：目的建立长循环红细胞载体中包载的地塞米松磷酸钠(DSP)含量测定方法。方法采用高效液相色谱法,色谱柱为Shimadzu Shim-pack VP-ODS色谱柱(250 mm×4.6 mm,5 μm),Waters Symmetry C18预柱(3.9 mm×22 mm,5 μm),内标为苯甲酸,流动相为甲醇-0.75%三乙胺溶液(60∶40,V/V),流速为1.0 ml/min,柱温为25℃,检测波长为242 nm。结果地塞米松磷酸钠在2.54~100.04 μg/ml浓度范围内与峰面积比值呈良好的线性关系,回收率>98%,供试溶液在4℃和室温下放置24 h均稳定,精密度符合要求。结论该法快速、简便、准确、重复性好,可用于长循环红细胞载体中地塞米松磷酸钠含量的测定。Objective To establish a high performance liquid chromatography(HPLC)method to determine the content of dexamethasone sodium phosphate(DSP)in long-circulating erythrocyte-based drugs. Methods Dexamethasone sodium phosphate was determined by HPLC with Shimadzu Shim-pack VP-ODS chromatographic column(4.6 mm×250 mm,5 μm)and Waters Symmetry C18 precolumn(3.9 mm×22 mm,5 μm). The internal standard was benzoic acid. The mobile phase was composed of methanol and 0.75% triethylamine solution at 60∶40(V/V)and the flow rate was 1.0 ml/min. The column temperature was 25℃ and the detection wavelength was 242 nm. Results The calibration curve of dexamethasone sodium phosphate showed a good linearity in the concentra? tion range of 2.54-100.04 μg/ml,and the average recovery was more than 98%. The test solution was stable both at 4℃ and room temperature for 24 h,and the precision met requirements. Conclusion The established HPLC method is simple,specific,accurate,repeatable and suitable for the determination of dexamethasone sodium phosphate in the long-circulating erythrocyte-based drugs.
