摘  要:目的探索中国农村汉族人群rs2880412、rs2880416和rs1047214位点基因多态性与肥胖易感性的关系。方法采用病例对照的研究方法选取农村地区410名肥胖患者和410名非肥胖者对照,抽取研究对象的静脉血10 ml提取DNA和进行体格检查,采用PCR-RFLP法进行基因多态性检测。结果两组间rs2880412位点基因型AA、CC和等位基因A、C分布差异均有统计学意义(P均<0.05),与等位基因A比较,C是肥胖的易感性基因,OR(95%CI)为1.29(1.06~1.56);rs2880416位点基因型CC、CG、CC+CG和等位基因C、G分布差异均有统计学意义(P均<0.05),与等位基因G相比,C是肥胖的易感性等位基因,OR(95%CI)为0.33(0.26~0.42)。肥胖的发病危险随着位点和等位基因突变个数的增加呈增加趋势(P均<0.05),且突变数量分别为3和4时能够增加肥胖易感风险,OR(95%CI)分别为2.01(1.03~3.94)和4.00(1.44~11.08)。结论肥胖遗传易感性与rs2880412和rs2880416位点基因多态性存在关联,对应等位基因C、C可能是肥胖发生的易感等位基因。Objective To explore the relationship between rs2880412,rs2880416,and rs1047214 gene polymorphism and obesity susceptibility in Chinese rural Han population.Methods Case control method was used to select 410 obese patients and 410 non-obese controls in a rural area.Venous blood of the subjects was extracted with 10 ml of DNA for physical examination,and PCR-RFLP method was used for genetic polymorphism detection.Results The distribution of AA,CC and allele A and C of rs2880412 between the two groups were statistically significant(all P<0.05).Compared with allele A,C was the susceptibility gene to obesity,and OR(95%CI)was 1.29(1.06~1.56).Rs2880416 genotype CC,CG,CC+CG and allele C and G distribution differences were statistically significant(all P<0.05).Compared with allele G,C was a susceptibility allele to obesity,and OR(95%CI)was 0.33(0.26~0.42).The risk of obesity increased with the increase of the number of loci and allele mutations(both P<0.05),and the number of mutations 3 and 4 could increase the risk of obesity susceptibility,with OR(95%CI)of 2.01(1.03~3.94)and 4.00(1.44~11.08),respectively.Conclusion The genetic susceptibility of obesity is associated with the polymorphism of rs2880412 and rs2880416 loci,and the corresponding alleles C and C may be the susceptible alleles of obesity.
