摘 要:目的探讨纳米TiO2对雌性小鼠的生殖毒性效应。方法将60只健康SPF级雌性ICR小鼠随机分为4组,分别为对照(生理盐水)组和10、50、100 mg/kg/d纳米TiO2染毒组,每组15只,连续28天灌胃染毒。检测小鼠生殖脏器系数、阴道涂片观察动情周期、HE染色观察卵巢组织变化、试剂盒测定卵巢中超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化物酶(POD)及过氧化氢酶(CAT)水平。结果染毒后,与对照相比,50和100 mg/kg/d染毒组,小鼠动情前期明显缩短(P<0.05),动情后期显著延长(P<0.05);卵巢组织HE切片观察发现,纳米TiO2染毒组中,小鼠卵巢颗粒细胞和黄体均出现不同程度的结构疏松和炎症细胞浸润;氧化应激测定结果表明,50和100 mg/kg/d染毒组小鼠卵巢SOD活性显著低于对照组(P<0.05);与对照相比,各染毒组MDA含量均显著增加(P<0.05);POD和CAT活性分别在100和50、100 mg/kg/d剂量组明显高于对照组(P<0.05)。结论纳米TiO2可引起小鼠动情周期紊乱及卵巢组织的氧化损伤。Objective To investigate the reproductive toxicity of nano titanium dioxide on female mice.Methods Sixty healthy female SPF ICR mice were divided into four groups randomly and averagely.Physiological saline was used as negative control,and different concentrations were used as experiment groups.Mice were administered via oral gavage in 0,10,50,or 100 mg/kg/d nano-TiO2 continuing 28 days.The organ coefficients were calculated and the estrous cycle was observed.And the histopathological changes of ovary were observed by HE staining,while the activity of SOD,POD,CAT and MDA content in ovary were analyzed by commercial kits.Results Compared with the control group,the proestrus cycle of mice treated with 50 and 100 mg/kg/d nano-TiO2 was significantly shorter(P<0.05),while the metestrus cycle was significantly longer(P<0.05).The SOD activity was significantly decreased in 50 and 100 mg/kg/d dose groups(P<0.05),while CAT activity was significantly increased(P<0.05).MDA content in all treated groups was significantly increased(P<0.05),and POD activity was significantly increased in the 100 mg/kg/d dose group(P<0.05).As for the exposure groups,the ovarian granulosa cells and corpus luteum were of loose structure and infiltration of inflammatory cells.Conclusion Nano titanium dioxide lead to estrous cycle disruption and ovarian injury when the female mice are exposed to nano dioxide.
