摘  要:2015年12月北京某养鸡场出现疑似禽流感疫情,采集病料接种鸡胚分离鉴定1株H9N2亚型禽流感病毒,命名为A/chicken/Huabei/2015(H9N2),通过RT-PCR扩增HA基因和NA基因,测序后进行了同源性分析,构建了HA基因遗传进化树,并对HA基因的受体结合位点、裂解位点和糖基化位点进行了分析。结果表明,分离株A/chicken/Huabei/2015 (H9N2)HA核苷酸序列与A/chicken/Jiangsu/J3588/2014(H9N2)的同源性最高(98.9%);NA核苷酸序列与A/chicken/Shandong/1167/2015(H9N2)的同源性最高(98.8%),遗传进化分析其属于欧亚分支中的Y280亚群。HA裂解位点为-PSRSSR/GLF-,符合低致病力毒株的特征;受体结合位点234位氨基酸为L,可能具有与SAα2,6Gal受体结合的特性;有8处潜在的糖基化位点。研究结果为H9N2亚型AIV的防控和疫苗研制提供了参考。In December 2015,a suspect avian influenza case occurred in a chicken farm in Beijing. One H9N2subtype avian influenza virus, designated as A/chicken/Huabei/2015(H9N2), was isolated by inoculating embryos with tissue samples. The HA gene and NA gene was amplified and sequenced. The nucleotide identity and genetic analysis of HA gene were carried out. We also analyzed the receptor binding sites,cleavage sites and glycosylation sites. The results showed that HA gene of A/chicken/ Huabei/2015(H9N2)had the highest nucleotide sequence identity of 98.9% with A/chicken/Jiangsu/J3588/2014 ( H9N2), and the NA gene had the highest nucleotide sequence identity of 98.8% with A/chicken/Shandong/1167/2015(H9N2). Phylogenetic tree of HA gene analysis showed that the isolate belonged to Eurasian branch of Y280 subgroup. The amino acid sequence at the cleavage region of the HA gene was -PSRSSR/GLF-, which is typical for low pathogenicity AIVs. The 234 amino acids at receptor binding site were L, which might had the characteristics of binding to SAα2, 6Gal receptors. There were 8 potential glycosylation sites on HA protein. The above research provided a scientific reference for the prevention and control of H9N2 subtype AIV and vaccine development.
