摘 要:目的研究化痰活血方(Huatan Huoxue Fang,HTHXF)对卵清蛋白(Ovalbumin,OVA)诱导的哮喘小鼠肺泡灌洗液(Bronchoalveolar lavage fluids,BALF)中趋化因子Eotaxin、IL-8、MCP-1及Th17细胞分化调控相关因子IL-1β、IL-21和IL-23生成的影响。方法雌性BALB/c小鼠48只随机分为正常组(C)、模型组(M)、地塞米松组(D)、化痰活血方低剂量组(HL)、化痰活血方中剂量组(HM)和化痰活血方高剂量组(HH)共6组,8只/组。各组于实验第1,8,15天腹腔注射OVA混悬液致敏(正常组注射生理盐水),第21~27天雾化吸入OVA诱导哮喘小鼠模型。于实验第21~27天雾化激发前1h灌胃给予各组药物。实验第28天收取BALF,采用ELISA法测定哮喘小鼠BALF中Eotaxin、IL-8、MCP-1、IL-1β、IL-21和IL-23的水平。结果化痰活血方能明显降低哮喘小鼠BALF中Eotaxin、IL-8、MCP-1、IL-1β、IL-21和IL-23的水平。结论化痰活血方治疗哮喘的作用机制可能与抑制趋化因子的分泌以及调控Th17型免疫反应有关。Objective To study the effects of Huatan Huoxue Fang(HTHXF)on the expression of chemokines Eotaxin,IL-8,MCP-1 and Th17 cell differentiation regulators IL-1β,IL-21,IL-23 in bronchoalveolar lavage fluids(BALF)of asthmatic mice induced by ovalbumin(OVA).Methods 48 female BALB/c mice were randomly divided into 6 groups(8 mice/group):normal group(C),model group(M),dexamethasone group(D),low dose group of Huatan Huoxue Fang(HL),middle dose group(HM)and high dose group(HH).The asthmatic mice were sensitized by intraperitoneal injection of OVA suspension on the 1st,8th and 15th day except for normal group(C),and the asthmatic mice model was induced by inhaling OVA stimulant on the 21st-27th day.The mice were given intragastric administration before 1h of atomization.BALF were obtained on the 28th day.The levels of Eotaxin,IL-8,MCP-1,IL-1β,IL-21,IL-23 in BALF were determined by ELISA.Results HTHXF could significantly reduce the levels of Eotaxin,IL-8,MCP-1,IL-1β,IL-21,IL-23 in BALF of asthmatic mice.Conclusion The mechanism of Huatan Huoxue Fang in the treatment of asthma is probably related to the inhibition of the secretion of chemokines and the regulation of Th17-type immune response.
