摘 要:目的通过比较绿原酸和体外血清外孵后的绿原酸对BN大鼠的致敏性差异,探讨绿原酸的致敏性及其作用机制,为临床合理用药提供参考。方法将绿原酸与大鼠血清进行孵育,对BN大鼠进行致敏和激发,比较孵育组与未孵育组动物在过敏反应发生率、过敏反应发生程度、激发前后血浆中组胺、类胰蛋白酶、β-氨基己糖苷酶、IgE含量及增长率;被动皮肤过敏试验大鼠皮下是否出现蓝斑以及蓝斑面积大小。结果(1)外孵绿原酸溶液组的过敏反应发生率、过敏反应的发生程度均高于绿原酸供试品溶液组;(2)绿原酸供试品溶液组激发前后血浆中组胺、类胰蛋白酶、β-氨基己糖苷酶、IgE的含量与生理盐水组比较无显著性差异;经体外孵育后,血浆中IgE、β-氨基己糖苷酶的含量均升高,与生理盐水组相比具有显著性的差异;激发前后四种指标的变化率均高于绿原酸未孵育组;(3)经体外孵育后,绿原酸溶液组被动皮肤过敏反应的阳性率为50.00%,绿原酸未孵育组被动皮肤过敏反应的阳性率仅为12.50%。结论绿原酸作为小分子物质,以半抗原形式存在,进入体内后能与血清蛋白结合形成复合物而产生免疫原性,从而引起过敏反应。Objective To promote clinical rational usage of chlorogenic acid(CA),the effect of CA and chlorogenic acid-BN rat serum on the allergenic reaction of BN rat were compared and its mechanism were investigated.Methods The artificial antigen was synthesized by conjugating Chlorogenic acid(CA)and the BN rats serum in vitro and sensitized samples in each group.Incidence rates,degree of allergenic reaction were observed,the difference between mixture groups and normal injection groups were compared by tryptase,histamine,β-hexosaminidase,IgE release and PCA test.R esults(1)The allergic rates of mixture groups are higher than CA groups and the allergic degree is more serious than that of CA groups.(2)IgE,histamine,tryptase andβ-hexosaminidase concentration of CA group with the corresponding normal saline group were no significant differences.Compared with normal saline group,the level of IgE andβ-hexosaminidase in the plasma of mixture group increased significantly and the release rate of IgE,histamine,tryptase,β-hexosaminidase in the plasma of mixture groups was higher than that in CA groups.(3)PCA test by using sera from mixture group after irritations showed the strong positive result characterized as large amount of subcutaneous effusions of Evans blue in SD rats and the rates of positive response are higher than that of CA groups.Conclusion CA is a kind of possible allergen which has strong allergenicity after combination with serum proteins.
