摘 要:目的从Neu3/IAP通路探讨竹节参总皂苷(saponins from Panax japonicus,SPJ)对衰老大鼠结肠炎症的改善作用及其可能的机制。方法SD雄性大鼠分为四组,青年组(6月龄)、自然衰老组(24月龄)以及竹节参总皂苷低、高剂量组。SPJ低、高剂量组大鼠从18月龄分别给予含10、30mg/kg的SPJ饲料至24月龄。测量结肠组织长度,生化试剂盒检测大鼠结肠组织中MPO水平,免疫组化检测炎症相关蛋白NLRP3、TNF-α的表达,以及结肠组织中紧密连接蛋白ZO-1、claudin-5的表达,酶组织化学染色法检测大鼠十二指肠IAP活性,RT-PCR检测结肠组织中IAP、Neu3mRNA表达。结果与自然衰老组比较,SPJ低、高剂量能显著降低大鼠结肠长度(P<0.05),增强MPO活力(P<0.05或P<0.01),降低炎症相关蛋白NLRP3、TNF-α(P<0.01),增加紧密连接蛋白ZO-1、claudin-5的表达(P<0.01或P<0.05),增强十二指肠IAP活性以及结肠组织中IAP、Neu3mRNA表达(P<0.01或P<0.05)。结论SPJ可能通过Neu3/IAP通路减轻肠道炎症反应。Objective To investigate the effects of saponins from Panax japonicus(SPJ)on colonic inflammation through Neu3/IAP signaling pathway in aging rats.Methods SD male rats were divided into four groups:the young group(6 months old),natural aging group(24 months old),and SPJ low-,high-dose groups.Rats of SPJ-treated groups had been treated with different doses(10 mg/kg,30 mg/kg)of SPJ since they were 18 months.The length of colon was measured.Levels of MPO were determined by biochemical kit.The expressions of NLRP3 and TNF-αwere determined by immunohistochemistry.The expressions of tight junction protein ZO-1 and claudin-5 were determined by immunohistochemistry.The expressions of IAP and Neu3 mRNA were determined by RT-PCR in the duodenum.Results SPJ decreased the colon length(P<0.05),increased MPO activity(P<0.01 or P<0.05),decreased the expression of inflammation-related proteins NLRP3 and TNF-α(P<0.01),increased expression of tight junction proteins ZO-1,claudin-5(P<0.01 or p<0.05),increased IAP activity of the duodenum and expression of IAP mRNA in colon(P<0.01 or P<0.05),decreased expression of Neu3 mRNA in colon(P<0.01).Conclusion The studies demonstrated that SPJ can inhibit inflammation in the colon of natural aging rats.The mechanism may be related to Neu3/IAP signaling pathway.
