摘  要:目的探讨急性肺损伤(Acute lung injury,ALI)后肺泡细胞的NIX基因及蛋白表达水平与创伤严重程度的关系及意义。方法建立不同严重程度大鼠胸部撞击伤模型,利用免疫组化、RT-PCR分子生物学技术、TUNEL分析法分别检测不同程度创伤模型大鼠,伤后6,12,24,48,72,96h肺泡细胞NIX基因及其蛋白表达水平。结果⑴创伤组与对照组均存在NIX蛋白表达,NIX在伤后6h开始表达增高,并在48h达到峰值,随后逐渐下降,96h后接近致伤前水平。⑵肺组织中NIX蛋白表达变化与肺损伤程度呈显著性正相关(r=0.303,P<0.01)。⑶致伤组与对照组肺泡相比细胞凋亡率显著增加(P<0.01)。结论NIX表达水平和肺损伤严重程度正相关,这可能是创伤后肺泡细胞损伤的分子生物学基础之一。Objective To investigate the relationship between the expression of NIX gene and protein in alveolar cells and the severity of trauma after acute lung injury (ALI).Methods A rat model of chest impact injury with different severity was established.The rats with different degrees of trauma were detected by immunohistochemistry, RT-PCR molecular biology technique and TUNEL analysis.After injury, 6, 12, 24, 48, 72, 96 h The expression level of NIX gene and its protein in alveolar cells.Results ⑴NIX protein expression was found in both the wound group and the control group.NIX began to increase at 6 h after injury and peaked at 48 h, then gradually decreased, and reached the pre-injury level after 96 h.⑵The expression of NIX protein in lung tissue was significantly positively correlated with the degree of lung injury (r=0.303, P<0.01).(⑶The apoptosis rate of the injured group was significantly higher than that of the control group (P<0.01).Conclusion The expression level of NIX is positively correlated with the severity of lung injury, which may be one of the molecular biological basis of post-traumatic alveolar cell injury.
