摘 要:利用同源克隆的方法设计特异性引物,从龙脑樟中获得一个333bp的基因片段,编码110个氨基酸。该片段的蛋白分子量为12.37ku,理论等电点为5.03,平均疏水性(GRAVY)为-0.312,α螺旋、β片层和无规则卷曲的比例分别为40.04%、10.09%和45.87%。同源比对和进化树分析结果都表明,龙脑樟Actin基因与香樟、山鸡椒的Actin基因具有较高的同源性。龙脑樟Actin基因片段的克隆,为深入研究该基因的表达调控及龙脑樟遗传与进化关系提供理论基础。A 333 bp gene fragment encoding 110 amino acids was obtained from Cirmamonun camphora by homologous cloning.The molecular weight was 12.37 ku,the theoretical pI was 5.03,and the grand average of hydropathicity (GRAVY) was -0.312.Moreover,the ratios of α helices,extended strand and random coil were 40.04%,10.09% and 45.87%,respectively.The results of homology comparison and phylogenetic tree analysis showed that the Actin gene of Cirmamonun camphora had high homology with Cirmamonun camphora and Capsicum capsicum.Cloning the Actin gene from Cirmamonun camphora was the basis for further study of the gene expression and evolutionary relationship.
