摘 要:目的研究结束艾灸治疗后1个月、2个月、3个月豚鼠血清对体外培养人黑素细胞增殖、黑素含量、酪氨酸酶活性的影响。方法将50只豚鼠随机分为5组,每组10只,分别为空白对照组、T-1组(艾灸治疗结束后即刻)、T-2组(艾灸治疗结束后1个月)、T-3组(艾灸治疗结束后2个月)、T-4组(艾灸治疗结束后3个月)。各组豚鼠背部脱毛约2cm×2cm,除空白对照组外,其余4组豚鼠均进行艾灸治疗,取穴为经外奇穴“灸癜风”和“阿是穴”。空白对照组仅采用同法固定,不进行艾灸。比较各组艾灸血清对体外培养人黑素细胞增殖、黑素含量、酪氨酸酶活性的影响。结果与空白对照组比较,T-1组的5%艾灸血清、T-2组的5%及10%艾灸血清均能够促进人黑素细胞增殖(P<0.05),T-2组5%艾灸血清的细胞增殖高于T-1组5%艾灸血清(P<0.05),T-2组5%艾灸血清的细胞增殖高于T-2组10%艾灸血清(P<0.05)。与空白对照组比较,T-1组、T-2组、T-3组和T-4组的艾灸血清均能够增加人黑素细胞中的黑素含量(P<0.05),T-2组的人黑素细胞中的黑素含量分别高于T-1组、T-3组和T-4组(P<0.05)。与空白对照组相比,T-2组、T-3组的艾灸血清可增强人黑素细胞中酪氨酸酶活性(P<0.05),T-2组的人黑素细胞中的酪氨酸酶活性高于T-3组(P<0.05)。结论艾灸血清可有效促进体外培养人黑素细胞增殖、黑素含量和酪氨酸酶活性,而且这种影响作用在艾灸治疗结束后3个月都存在,尤其在结束艾灸治疗后1个月的效果最为明显。Objective To study the effect of guinea pig moxibustion serum on the proliferation,melanin content and tyrosinase activity of human melanocytes cultured in vitro at 1 month,2 months and 3 months after moxibustion treatment.Method Fifty guinea pigs were randomized into 5 groups,with 10 in each group,including blank control group,T-1 group (0 month after moxibustion),T-2 group (1 month after moxibustion treatment),T-3 group (2 months after moxibustion treatment) and T-4 group (3 months after moxibustion treatment).The back of all the guinea pigs was shaved by approximately 2 cm×2 cm.Except for the blank control group,the other four groups of guinea pigs were treated with moxibustion at Jiudianfeng and Ashi points.The blank control group was only fixed by the same method but not given moxibustion.The effects of moxibustion serum on the proliferation,melanin content and tyrosinase activity of human melanocytes cultured in vitro were compared.Result Compared with the blank control group,5% moxibustion serum in the T-1 group,and 5% and 10% moxibustion serum in the T-2 group promoted the proliferation of human melanocytes (P<0.05);the proliferation led by 5% moxibustion serum in the T-2 group was more significant than that caused by 5% moxibustion serum in the T-1 group (P<0.05) and 10% moxibustion serum in the T-2 group (P<0.05).Compared with the blank control group,moxibustion serum of the T-1,T-2,T-3 and T-4 groups increased the melanin content in human melanocytes (P<0.05),and the melanin content in the T-2 group was higher than that in the T-1,T-3 and T-4 groups (P<0.05).Compared with the blank control group,moxibustion serum in the T-2 group and T-3 group enhanced the tyrosinase activity in human melanocytes (P<0.05),and the tyrosinase activity in the T-2 group was higher than that in the T-3 group (P<0.05).Conclusion Moxibustion serum can effectively promote the proliferation,increase the melanin content and tyrosinase activity of human melanocytes cultured in vitro.This effect even still exists three months aft
