摘 要:以红车轴草提取物为试材,乳酸脱氢酶、黄嘌呤氧化酶、α-葡萄糖苷酶作为生物靶分子,采用"受体-配体"亲和超滤法、超高效液相色谱-高分辨质谱联用技术(UPLC-Q-Exactive)、半制备型液相色谱法,筛选、鉴定、分离红车轴草提取物中具有酶抑制作用的活性配体,以期治疗脑卒中病和糖尿病。结果表明:红车轴草提取物中有6种具有酶抑制作用的活性配体,即大豆苷、芒柄花苷、大豆黄素、染料木素、芒柄花素和鹰嘴豆芽素A,通过半制备型液相色谱分离的6种活性配体纯度均高于92%,具有治疗脑卒中病和糖尿病的潜在价值。Trifolium pratense extracts were used as test materials,and lactate dehydrogenase(LDH),xanthineoxidase(XOD) and α-glucosidase were used as biological target molecules.In order to treat stroke and diabetes,the method of receptor-ligand affinity ultrafiltration,ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry(UPLC-Q-Exactive) and semi-preparative liquid chromatography were used to screen active ligands with enzyme inhibition in Trifolium pratense extracts.The results showed that there were six active ligands with enzyme inhibition in the extracts of Trifolium pratens,namely:daidzin,ononin,daidzein,genistein,formononetin and biochanin A.The purity of the six active ligands were separated by semi-preparative liquid chromatography was above 92%,which has potential value in the treatment of stroke and diabetes.
