摘 要:目的探讨丹参川芎嗪注射液对急性肺栓塞(APE)大鼠血清生长分化因子-15(GDF-15)及氨基末端脑钠肽前体(NT-proBNP)的影响及意义。方法健康雄性Wistar大鼠64只,随机分为7天组(n=32)和7小时组(n=32),每组大鼠又随机分为栓塞组(n=8)、肝素组(n=8)、丹参川芎嗪组(n=8)和联合用药组(n=8)。通过异体血栓输入复制APE模型。分别于注栓后7天、7小时取血,离心后通过酶联免疫吸附试验检测血清GDF-15及NT-proBNP的浓度并进行分析。结果肝素组、丹参川芎嗪组、联合用药组于各时间段血清GDF-15、NT-proBNP浓度较栓塞组均有降低,但程度不同,其中联合用药组于7小时血清GDF-15、NT-proBNP浓度较栓塞组改变有显著意义(P<0.05)。结论应用丹参川芎嗪注射液后急性肺栓塞大鼠相较于未应用的大鼠血清生长分化因子-15及NT-proBNP均有不同程度的降低,尤其联合低分子肝素进行治疗,两项生物标记物降低更为显著。Objective To investigate the effect of Salvia miltiorrhiza Ligustrazine injection on serum growth differentiation factor-15(GDF-15)and precursor of amino-terminal brain natriuretic peptide(NT-proBNP)in rats with acute pulmonary embolism(APE).Methods There were 64 healthy male Wistar rats,and they were randomly divided into the 7-day group(n=32)and the 7-hour group(n=32).Each group was randomly divided into the embolization group(n=8),the heparin group(n=8),the Salvia miltiorrhiza Ligustrazine group(n=8)and the combination therapy group(n=8).The APE model was replicated through allogenic thrombus,and their blood was collected 7 days and 7 hours after injection respectively.After centrifugation,serum GDF-15 and NT-proBNP concentrations were detected by enzyme-linked immunosorbent assay(ELISA).Results The concentrations of GDF-15 and NT-proBNP in serum of the heparin group,the Salvia miltiorrhiza Ligustrazine group and the combined drug group were lower than those of the embolization group,but the degree was different.The levels of GDF-15 and NT-proBNP in the combined group were significantly higher than those in the embolization group at 7 hours(P<0.05).Conclusion The levels of serum growth differentiation factor-15 and NT-proBNP in acute pulmonary embolism rats treated with Salvia miltiorrhiza and ligustrazine injection are lower than those in untreated rats,especially in combination with low molecular weight heparin.
