摘 要:以琼脂糖凝胶介质Bestarose CL 4B为基质,苄胺为配基,制备葡聚糖接枝型混合模式层析介质.考察葡聚糖分子量对接枝量,及接枝过程对微球形貌、粒径分布、物理性质和蛋白吸附性能的影响.结果表明,葡聚糖接枝量与其分子量成正比,接枝过程对微球的形貌及粒径分布基本无影响,葡聚糖接枝后微球的湿球密度基本不变,含水率和孔体积降低.接枝介质BA-4B-T40对BSA的饱和吸附容量比未接枝介质提高约22.5%,对IgG的吸附无明显效果.Dextran-grafted mixed-mode chromatography resins were prepared by using particle Bestarose CL 4 B as the matrix and benzylamine as the ligand. The effect of dextran molecular weight on the amount of dextran grafted and effect of grafting process on the particle morphology, particle size distribution, physical properties and protein adsorption performance were studied. The results demonstrated that the amount of dextran grafted increased with increasing dextran molecular weight. After dextran grafting, the particle morphology and particle size distribution were almost unchanged, while water content and pore volume decreased. Compared with non-grafting resin, dextran-grafted resin BA-4 B-T40 showed 22.5% improvement in saturated adsorption capacity towards BSA. While the adsorption performances of dextran-grafted resins against IgG did not change obviously.
