摘 要:为评价免疫增强剂CVC1302对猪口蹄疫疫苗的免疫增强效力,本研究以50日龄断奶仔猪为研究对象,将不同浓度的CVC1302添加到口蹄疫疫苗中免疫仔猪,免疫后28 d采血,液相阻断ELISA (LPB-ELISA)检测免疫猪抗体水平,以筛选CVC1302最佳使用剂量;采用LPB-ELISA监测最佳使用剂量下疫苗对免疫仔猪的免疫持续期;免疫后28 d对所有猪以1000倍半数感染量(PID50) O型口蹄疫病毒(FMDV) MYA98株攻毒评价疫苗的保护效力。同时评价免疫增强剂CVC1302对现有商品化疫苗(O型灭活疫苗、O/A/Aisa-1型三价灭活疫苗及合成肽疫苗)的免疫增强效力。结果表明,高、中、低3个剂量的CVC1302均能够极显著提高口蹄疫疫苗的特异性抗体水平约3个滴度(p<0.01),并且中剂量的CVC1302效果最佳。CVC1302可以延长口蹄疫疫苗的合格抗体持续期达6个月,并可以将灭活疫苗的攻毒保护力(PD50)从6.9提升至15.59。将CVC1302添加到商品化疫苗中能够极显著提高口蹄疫疫苗的免疫效力(p<0.01)。结果表明,免疫增强剂可以较大幅度增强口蹄疫疫苗的免疫效力,具有较为广阔的应用前景。本研究为提高口蹄疫疫苗的免疫效果提供了新的选择。In order to further evaluate the efficiency of immunopotentiator CVC1302, 50-days piglets were vaccinated with or without adding a of CVC1302 to the FMD vaccine at three different doses and the serum antibody were detected by LPB-ELISA at 28 days post vaccination for screening optimal dose of cvc1302. Then, antibody duration of optimal dose group of cvc1302 were measured. Finally, all pigs wre immuned with different doses FMDV and all piglets were challenged with 1000-fold PID50 FMD virus to measure the protection rate at 28 days post vaccination. In addition, CVC1302 was applied to various commercial vaccines(inactivated vaccines, inactivated trivalent vaccines and peptide vaccines) to investigate the immune enhancement effect. Results showed that all of the high, medium or low dose of CVC1302 can remarkably increase the specific antibody titers by three times,and medium dose was the best one. The immunity of the vaccine adjuvant with CVC1302 lasted for 6 months with PD50 improved from 6.9 to 15.59. CVC1302 also can significantly improve the specific antibody of commercial FMD vaccines. This study provides a new option for improving the immune effect of foot and mouth disease vaccine.
