摘 要:目的探讨加味芎蝎散对咳嗽变异性哮喘(CVA)大鼠的作用及上皮连接蛋白表达的影响。方法SD大鼠40只,随机分为空白对照组10只,造模组30只。肌注卵蛋白联合腹腔注射Al(OH)3法建立CVA大鼠模型。随后将30只造模成功大鼠随机分为模型对照组、孟鲁司特钠组、中药组,每组10只。对照组、模型组均灌服蒸馏水;孟鲁司特钠组10mg/kg,中药灌服50g/kg,每天1次;灌药4周后处死大鼠,取肺组织标本,ELISA法检测气道组织中细胞因子白介素-33(IL-33)和胸腺基质淋巴细胞生成素(TSLP)的表达,光镜下观察支气管组织形态,免疫组化及Western blotting法检测各组支气管黏膜上皮细胞连接蛋白E-caderin、ZO-1以及Claudin3的表达。结果与对照组比较,模型组大鼠肺组织内细胞因子IL-33和TSLP含量明显升高,同时,肺组织内细胞连接蛋白E-caderin、ZO-1以及Claudin3表达下调。与模型组比较,中药组IL-33和TSLP含量减低,Ecaderin、ZO-1以及Claudin3表达增高。结论加味芎蝎散汤可减轻CVA大鼠Th2型炎症反应,其机制可能与增强细胞间连接蛋白的表达,缓解气管黏膜上皮细胞损伤有关。Objective:To explore the effect of Jiawei Xiongxie Decoction on expression of epithelial junction-related proteins in rats with cough variant asthma.Methods:40 SD rats were randomly divided into the blank control group(10 rats)and the model group(30 rats).30 successful cough variant asthma rat models were randomly divided into the model control group,montelukast sodium group and traditional Chinese medicine group,10 rats in each.The control group and the model group were fed with distilled water;the other groups took corresponding drugs,once a day.After 4 weeks of administration,rats were sacrificed and lung tissue specimens were taken.The expression of cytokines IL-33 and TSLP in airway tissue was detected by ELISA,and the morphology of bronchial tissue was observed under light microscope.The expression of connexin E-caderin,ZO-1 and Claudin-3 in bronchial epithelial cells was detected by immunohistochemistry and Western blotting.Results:Compared with the control group,the levels of cytokines IL-33 and TSLP in lung tissue of CVA rats were significantly increased, while the expressions of connexin E-caderin,ZO-1 and Claudin 3 in lung tissue were down-regulated;compared with the model group,the contents of IL-33 and TSLP decreased and the expressions of E-caderin,ZO-1 and Claudin-3 increased in the traditional Chinese medicine group.Conclusion:Jiawei Xiongxie Decoction can alleviate TH2 inflammation in CVA rats.Its mechanism may be related to enhancing the expression of connexin and alleviating the injury of tracheal epithelial cells.
