摘 要:为实现蘘荷[Zingiber mioga(Thunberg)Roscoe]的离体快速繁殖,提高繁殖率,缩短培养周期,本试验以蘘荷成熟种子萌发出的幼苗作为外植体,以Murashige and Skoog(MS)为基本培养基,添加不同浓度的6-苄基腺嘌呤(6-Benzylaminopurine,6-BA)、激动素(Kinetin,KT)、噻苯隆(Thidiazuron,TDZ)和萘乙酸(1-Naphthylacetic acid,NAA),探究这些植物生长调节剂对蘘荷芽增殖倍数及一步成苗的影响.结果表明,低浓度TDZ对蘘荷丛生芽增殖影响显著,以添加0.01 mg/L TDZ的增殖效果最优,丛生芽增殖倍数为9.0;6-BA次之,在添加2.0 mg/L 6-BA时,丛生芽增殖倍数最高,为7.28;KT效果最差,在添加3.0 mg/L KT时,丛生芽增殖倍数最高时仅为4.5.然而,在丛生芽增殖倍数高的MS+0.01 mg/L TDZ培养基上,其生根率(38%)、平均株高(2.28 cm)、单株平均根数(0.80)及平均根长(0.52 cm)均较低,所获得的再生植株不适宜移栽.而在添加MS+2.0 mg/L 6-BA的增殖培养基上,再生植株平均株高5.20 cm、生根率100%、单株平均根数9.20,平均根长4.08 cm,株形好,不经生根培养即可进行移栽.通过本研究得到满足较高增殖率且同时生根后适宜移栽的一步成苗培养基为MS+3.0~4.0 mg/L 6-BA+0.01 mg/L NAA及MS+2.0~4.0 mg/L 6-BA.The seedlings germinated from mature seeds of Zingiber mioga (Thunberg) Roscoe were used as explants and the Murashige and Skoog (MS) was employed as base medium supplemented with different concentration of 6-Benzylaminopurine (6-BA), Kinetin (KT), Thidiazuron (TDZ), and 1-naphthylacetic acid (NAA) to study the effects of these plant growth regulators (PGRs) on shoot proliferation times and one-step seedling formation. The result showed that low concentration TDZ (0.01 mg/L) had best effect on shoot multiplication, the shoot proliferation rate was 9.0. The most suitable 6-BA concentration was 2.0 mg/L, the shoot proliferation rate was 7.28. KT had the worst multiplication effect, and the highest proliferation rate was 4.50 on MS+3.0 mg/L KT. However, although MS+0.01mg/l TDZ are best for shoot multiplication, low rooting rate(38%), mean shoot length (2.28 cm), mean rooting number (0.80) and mean root length (0.52 cm) were obtained in this medium, the regenerated seedlings from the medium failed to meet requirement of acclimatization. MS+2.0 mg/L 6-BA exhibited good growth condition among all growing indexes such as mean shoot length (5.20 cm), rooting rate (100%), mean root number (9.20) and mean root length (4.08cm). MS+2.0-4.0 mg/L 6-BA and MS+3.0-4.0 mg/L 6-BA+0.01 mg/L NAA were suitable for one-step seedling formation.
