摘 要:为了筛选适合鉴定甜瓜种子纯度的SSR标记,本研究以18份厚皮甜瓜品种组合为研究材料,从73对SSR (Simple sequence repeats)引物中筛选出适合各自品种纯度鉴定的SSR引物,并结合大田传统鉴定方法对其种子纯度进行了对比分析。结果表明,适合‘西州密1号’、‘西州密3号’、‘西州密25号’、‘西州密17号’、‘西州密21号’和‘西州密29号’种子纯度鉴定的具有双显性且条带清晰SSR引物分别有4对、4对、10对、9对、3对和2对;引物MU5499可作为以上品种纯度鉴定的通用引物。选择引物C30用于‘西州密25号’F1代种子纯度鉴定,使用该标记共检测191株,其中4株为异株,种子纯度为97.91%。试验用的‘西州密25号’种子田间鉴定纯度为98.99%,鉴定结果与SSR标记鉴定结果一致。SSR分子标记方法更能准确鉴别出甜瓜杂种一代中的不纯株,且大大缩短纯度鉴定周期,为西州密系列甜瓜杂交种纯度室内快速检测提供技术支撑。The aims of this study were to screen simple sequence repeat(SSR) markers for indentification of 18 Cucumis melon ssp. melo varieties, including six F1 hybrids(’Xizhoumi1’,’Xizhoumi3’,’Xizhoumi25’,’Xizhoumi17’,’Xizhoumi21’,’Xizhoumi29’) and their parents. Several pairs of specific primers out of 73 pairs of SSR primers were selected to make a rapid variety purity test, and the seed purity was compared and analyzed by combining with the traditional field identification methods. The results showed that the SSR primers with double dominant and clear bands were 4 pairs, 4 pairs, 10 pairs, 9 pairs, 3 pairs and 2 pairs, respectively, which were suitable for the identification of seed purity of ’Xizhoumi1’,’Xizhoumi3’,’Xizhoumi25’,’Xizhoumi17’,’Xizhoumi21’ and’Xizhoumi29’. The primer MU5499 could be used as a universal primer for purity identification of above varieties.The primer C30 was used to identify seed purity of the F1 generation of ’Xizhoumi 25’, a total of 191 plants were detected, four of which were miscellaneous varieties, and the seed purity was 97.91%. It was 98.99% that the field identification purity of seeds of ’Xizhoumi25’ used in the test, and the identification results were consistent with those of SSR marker identification results. The SSR marker identification method can more accurately identify the impure plants in the hybrid generation of muskmelon, and shorten the appraisal period greatly, which took technical support for the requirements of rapid variety purity test of the muskmelon new hybrids from Xizhoumi series.
