摘  要:为掌握百合茎尖培养脱毒技术,为百合脱毒苗的大批量生产提供技术支撑,以百合品种卡瓦娜(Corvara)为材料,对不同来源的培养材料(未经处理的组培苗、经过4℃低温处理40d后的组培苗和商品球)进行茎尖观察,并对适合培养的茎尖材料进行不同配方的组织培养。结果表明:未经处理的组培苗其茎尖生长点未见叶原基,其分生组织不具备立刻发育成茎的能力,不适宜作茎尖培养材料;经过低温处理的组培苗部分茎尖为完整的芽,顶端生长点活动旺盛,适宜进行茎尖培养,部分则无叶原基等其他结构;商品球茎尖部位明显且生长旺盛,最适宜作茎尖培养材料。百合茎尖组织培养的最佳培养基配方为MS+6-BA1.0mg/L+NAA0.5mg/L+蔗糖30g/L+琼脂7g/L+活性炭1g/L,pH5.8。To master the detoxification technology of lily shoot tip culture,and to provide technical support for mass production of virus-free lily seedlings,using Corvara as the material,the shoot tips of different sources(untreated tissue culture seedlings,tissue culture seedlings and commercial bulbs treated at 4℃for 40 days)were observed.Meanwhile,the stem tip materials suitable for culture were cultured with different formulations.Results:The shoot tip growth point of untreated tissue culture seedlings did not have leaf primordia,and its meristem did not have the ability to develop into stem immediately,so it was not suitable to be used as stem tip culture material;Nevertheless,some of the shoot tips of tissue culture seedlings treated with low temperature were complete buds,and the apical growing point was active,which was suitable for shoot tip culture,and some of them had no leaf primordia and other structures;Meanwhile,commercial bulb tip was obvious with vigorous growth,so it was the most suitable material for stem tip culture.The optimum medium for shoot tip tissue culture of lily was MS+6-BA 1.0 mg/L+NAA0.5 mg/L+sucrose 30 g/L+agar 7 g/L+activated carbon 1 g/L,pH 5.8.
