一个木聚糖降解菌群的分离及木聚糖酶酶学性质分析

摘  要:为从环境中分离出能够降解木聚糖的微生物,利用富集培养法从猪粪与甘蔗叶混合发酵堆肥中分离到一个具有木聚糖降解能力,能够利用玉米芯作为惟一碳源的混合菌群。高通量测序分析发现该菌群具有丰富的微生物多样性,其中未知微生物的含量约为50.8%。该菌群发酵液中的木聚糖酶在以AZO-xylan (Birchwood)为底物时,其最适反应温度为55℃,最适反应pH为8.0,在55℃以下能够保持很好的稳定性。重要的是,该菌群的发酵液能够直接水解玉米芯生产木三糖和木四糖,从而省去了碱处理玉米芯制备木聚糖的步骤,进而降低了环境污染以及生产成本。In this study a xylan-degrading bacterial community was isolated from pig manure and sugarcane leaf composting fermentation using enrichment culture method. The bacterial community was able to degrade xylan, and used corncob powder as the sole carbon source. High-throughput sequencing analysis revealed that the bacterial community had rich microbial diversity and the unknown microbial content was about 50.8%. Xylanase in the fermentation broth of the bacterial community reached the highest activity at temperature of 55 ℃ and pH of 8.0,and the enzyme showed very good thermal stability below 55 ℃. The most important thing was that the enzyme can directly hydrolyze corncob to get xylotriose and xylotetraose, eliminating the process of preparing xylan from corn kernels by alkali treatment and reduce environmental pollution and production cost.
