摘 要:目的观察大鼠在正常盐饮食情况下,醛固酮增多时对主动脉和尾加压素Ⅱ(UⅡ)及其受体(UT)的影响。方法将16只大鼠随机分为正常组和醛固酮组,每组8只,醛固酮组采用醛固酮皮下注射(20μg/d),常规饲养6周,用Masson染色血管胶原沉积情况,用放射免疫法检测血浆和主动脉组织UⅡ的含量;用免疫组织化学法检测主动脉组织UT的表达。结果Masson染色可见醛固酮组大鼠主动脉有大量胶原沉积,主动脉组织UⅡ含量明显上调(P<0.01),UT表达同步上调,但是两组血浆UⅡ含量比较差异无统计学意义(P>0.05)。结论醛固酮可上调UⅡ及其受体表达,并促进动脉纤维化,提示醛固酮上调UⅡ及其受体表达可能是其促进动脉纤维化的另一作用途径。Objective To investigate the effect of excess aldosterone on the aorta and UrotensinⅡ(UⅡ)and it′s receptor(UT)of rats with normal salted diet.Methods Sixteen SD rats were randomly divided into normal group(n=8)and aldosterone group(n=8).The rats in aldosterone group were infused subcutaneously with aldosterone(20μg/d)for 6 weeks.The expression of collagen was determined by Masson staining,the contents of UⅡin plasma and aorta were determined by radioimmunoassay,and the expressions of UT was determined by immunohistochemistry.Results Masson staining showed that there was mass collagen deposited in aorta of rats in aldosterone group,the expression of UⅡand UT on aorta were up-regulated significantly(P<0.01).There was no significantly differences in plasma UⅡcontents between two groups(P>0.05).Conclusion Aldosterone up-regulated the expression of urotensinⅡand it′s receptor,promoted vascular fibrosis.The result suggesed that aldosterone up-regulated the expression of urotensinⅡand it′s receptor might be another pathway to promote arterial fibrosis.
