摘  要:目的研究西红花苷是否能通过转化生长因子β1(TGF-β1)和核因子-κB(NF-κB)通路抑制草酸引起的肾小管细胞上皮间质转化(EMT)的发生。方法MDCK培养后分为3组(空白对照组、草酸组、草酸+西红花苷组)分别予以相应干预。运用Western blot蛋白印迹分析检测E-钙黏附蛋白(E-cadherin)、神经-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)、TGF-β1、NF-κB的表达。分别检测不同组细胞中丙二醛(MDA)、超氧化物歧化酶(SOD)、活性氧(ROS)的表达。结果草酸会使TGF-β1、Ncadherin、Vimentin、NF-κB表达量增加,使E-cadherin表达下降。西红花苷会降低草酸引起的细胞ROS的增加。结论西红花苷通过TGF-β1和NF-κB通路抑制上皮间质转化。Objective To investigate whether crocin can inhibit oxalate-induced epithelial-mesenchymal transition(EMT)of renal tubular epithelial cells via TGF-β1 and NF-kB signaling pathways.Methods MDCK cells were divided into three groups:control group,oxalate group and crocin+oxalate group.The protein expressions of E-cadherin,N-cadherin,Vimentin,transforming growth factorβ1(TGF-β1)and nuclear factor-κB(NF-κB)were detected with Western blot.The levels of malondialdehyde(MDA),superoxide dismutase(SOD)and reactive oxygen species(ROS)were determined.Results Oxalate treatment increased the levels of TGF-β1,N-cadherin,Vimentin and NF-κB while decreased the expression of E-cadherin.The addition of crocin markedly reduced the level of ROS.Conclusion Crocin can inhibit oxalate-induced EMT of renal tubular epithelial cells through TGF-β1 and NF-κB pathways.
