摘 要:目的研究茵陈蒿汤对非酒精性脂肪肝(nonalcoholic fatty liver disease,NAFLD)伴糖尿病(diabetes mellitus,DM)大鼠模型p38丝裂原活化蛋白激酶(p38MAPK)表达的影响。方法将SD大鼠随机分为对照组、DM组和DM+药物组,对照组予以常规饮食,而DM组和DM+药物组均采用高脂饮食,建立NAFLD合并DM大鼠模型,DM+药物组予以茵陈蒿汤。观察3组大鼠的体质量、肝重、肝体比、空腹血糖、生化指标[丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、胆固醇(TC)]以及肝脏组织p38MAPK的表达改变。结果 DM组大鼠的体质量显著高于对照组(P <0.05),而DM+药物组大鼠的体质量则显著降低(P <0.05)。DM组大鼠的肝重及肝体比均显著高于对照组(P <0.05),而DM+药物组大鼠的肝重及肝体比均显著降低(P <0.05)。DM+药物组大鼠的空腹血糖水平亦较DM组明显降低(P <0.05)。DM组大鼠的ALT、AST和TC水平均较对照组显著升高(P <0.05);而与DM组相比,DM+药物组大鼠ALT、AST和TC水平均显著降低(P <0.05)。DM组大鼠肝脏p38MAPK表达水平显著低于对照组(P<0.05),而DM+药物组的p38MAPK表达水平显著高于DM组(P <0.05)。结论茵陈蒿汤通过调控p38MAPK表达水平减轻NAFLD合并DM大鼠的肝脏损害和糖代谢异常。Objective To explore the effect of Yinchenhao decoction on p38 mitogen-activated protein kinase(p38MAPK)expression of nonalcoholic fatty liver disease(NAFLD)and diabetes mellitus(DM)in mice.Methods SD mice were randomly distributed into three groups:the control group,DM group and DM + drug group. Mice in the control group were fed regularly and those in the DM group and DM + drug group were fed with high fat diet. DM + drug group was also treated with Yinchenhao decoction. The body weight,fasting blood glucose,and other biochemical biomarkers(ALT,AST,TC)as well as p38MAPK expression in liver of the mice were measured and compared among three groups. Results The body weight in the DM group was much higher than the control group(P < 0.05),but the body weight of DM + drug group showed significantly lower than the DM group(P < 0.05). The liver weight and the ratio between liver and body weight were much higher in the DM group than the control group(P < 0.05),though they were much lower in the DM + drug group(P < 0.05). The value of fasting blood glucose in the DM + drug group was obviously decreased when compared to the DM group(P < 0.05). Mice in the DM group showed significant higher level of ALT,AST and TC than in the control group(P < 0.05). When compared to the DM group,mice in the DM + drug group showed much lower expression level of ALT,AST and TC(P < 0.05). The expression level of p38MAPK in the livers in the DM group was significantly lower than that in the control group(P < 0.05),and it was obviously increased in the DM + drug group(P < 0.05).Conclusion Yinchenhao decoction can alleviate liver damage and abnormal glucose metabolism in mice with NAFLD and DM by regulating the expression of p38MAPK.
