益气养阴方含药血清联合化疗药物对KG1α细胞增殖的影响

摘　　要：目的:观察益气养阴方含药血清联合化疗药物(阿糖胞苷、喜树碱、紫杉醇)对KG1a细胞增殖作用的影响。方法:予Wistar大鼠中药制剂灌胃制备含药血清;实验分为空白血清组,5%、10%、15%、20%含药血清组,化疗药单用组,含药血清联合化疗药组;各组细胞培养12、24、48、72 h,采用CCK8检测KG1a细胞增殖抑制率。结果:15%含药血清组12 h对KG1α细胞增殖抑制效果最为明显,随着时间的推移,抑制效果稍有所减弱,与空白血清组比较差异具有统计学意义(P <0. 01)。阿糖胞苷、紫杉醇对于KG1α细胞的抑制效果不明显;但在含药血清的协同作用下,对KG1α细胞的抑制作用明显增加,12 h即表现出明显的抑制作用,与空白血清组及单用化疗药组比较有显著统计学意义(P <0. 01)。喜树碱对KG1α细胞的抑制作用明显,并随着时间增加而增强;在含药血清的作用下,抑制效果进一步增强。结论:中药益气养阴方含药血清可增加KG1α细胞对化疗药的敏感性,增加细胞生长的抑制率。Objective: To observe the effects of drug serum containing Yiqi Yangyin Fang combined with chemotherapeutic drugs( cytarabine,camptothecin,paclitaxel) on proliferation of KG1α cells. Methods: The drug serum containing of Yiqi Yangyin Fang was prepared by Wistar rats. Experiment was divided into blank serum group,5%,10%,15%,20% drug-containing serum group,single chemotherapy drug group,drug-containing serum combined with chemotherapy drug group. KG1 a cells in each group was cultured for 12,24,48,72 hours. The inhibitory rate of KG1 a cells proliferation was determined by CCK8. Results: The inhibitory effect of 15% drug serum containing Yiqi Yangyin Fang on KG1 a cells was significant at 12 hour compared with the blank serum( P < 0. 01). The inhibitory effect of cytarabine and paclitaxel alone on KG1 a cells was not obvious,however,with drug-containing serum,the inhibition was significantly increased since 12 hours. The inhibitory effect of camptothecin on KG1 a cells is obvious and increased with the cultured time,under the action of drug-containing serum,it was more obvious. Conclusion: The drug serum containing Yiqi Yangyin Fang could enhance the sensitivity of KG1α cells to chemotherapeutic drugs and increase the inhibition rate of cells.
