森登-4味汤对膝骨性关节炎家兔关节滑膜组织IL-1、TNF-α和TGF-β水平变化的影响

摘　　要：目的:探讨蒙药森登-4味汤对家兔膝骨性关节炎的作用机制。方法:实验分为正常组、模型组和森登-4味汤高、中、低剂量组,每组16只家兔。改良Hulth法建立膝关节骨性关节炎家兔模型。各组分别给与相应处理28 d后,取家兔膝关节滑膜组织,ELISA检测滑膜组织白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)、转化生长因子β(TGF-β)等细胞因子水平。结果:模型组家兔膝关节滑膜组织IL-1β、和TNF-α水平明显高于正常组,组间比较有显著性差异(P <0. 01);森登-4味汤组家兔膝关节滑膜组织IL-1β、和TNF-α水平均明显低于模型组,差异有统计学意义(P <0. 05或P <0. 01);模型组家兔膝关节滑膜组织TGF-β水平明显低于正常组,组间差异有显著统计学意义(P <0. 01);森登-4味汤组家兔膝关节滑膜组织TGF-β水平均上调,差异有统计学意义(P <0. 05或P <0. 01)。结论:蒙药森登-4味汤能通过抑制IL-1β及TNF-α、上调TGF-β的分泌,有效减轻膝骨性关节炎家兔的炎症。Objective: To explore the mechanism of Mongolian Senden-4 Wei Tang on knee osteoarthritis.Methods: Experiment was divided into normal,model and Senden-4 Wei Tang high,middle,low dose groups,each group with 16 rabbits. Models of knee osteoarthritis were made by modified Hulth method. Each group was given the corresponding distilled water or different concentration Senden-4 Wei Tang respectively for 28 days,taking the synovia tissue of rabbits’ knee joints,IL-1β,TNF-αand TGF-βlevels in synovia tissue were determined by ELISA. Results: The IL-1β,TNF-α levels in synovia tissue of knee joints in model group were obviously higher than those of normal group( P < 0. 01),which in Senden-4 Wei Tang groups were significantly lower than those in model group,the differences were statistical significant( P < 0. 05,P < 0. 01);TGF-βlevels in synovia tissue of knee joints in model group was lower than that in normal group( P < 0. 01),that in Senden-4 Wei Tang groups was raised significantly,he differences were statistical significant( P < 0. 05,P < 0. 01). Conclusion: Mongolian Senden-4 Wei Tang can inhibit IL-1β,TNF-α and promote TGF-βsecretion,alleviate the inflammation of rabbits with knee osteoarthritis.
