摘  要:以湖北海棠1年生实生苗为研究对象,研究了淹水胁迫对其叶片生理指标的影响。结果表明:随着淹水时间延长,湖北海棠叶片可溶性糖、可溶性蛋白质量分数均显著高于对照组,并呈现逐步上升趋势;超氧化物歧化酶(SOD)活性先上升后下降,过氧化物酶(POD)先下降后上升,二者可能存在一定的补偿机制;脯氨酸(PRO)和丙二醛(MDA)含量均呈现先下降后上升的趋势。除可溶性糖外,其他生理指标的峰谷值出现的时间均为第10d,但各指标的启动时间不同,PRO与MDA在0~5d启动,SOD与POD以及可溶性糖在5~10d启动,可溶性蛋白在15~20d启动。通过荧光定量PCR检测水淹胁迫后胁迫相关基因的表达,初步认为MhWRKY1、MhWRKY40b和MhMYB121基因可能参与了湖北海棠响应淹水胁迫的响应。We studied the effects of waterlogging stress on the change of physiological indexes of Malus hupehensis leaves to explore the regulation and to evaluate the characteristics of waterlogging resistance for the cultivation of M. hupehensis . The contents of soluble sugar and soluble protein in leaves of M. hupehensis significantly increased than the control group and the contents showed an obvious escalating trend. The activity of T-SOD increased first and then decreased. Oppositely, the POD decreased first and then increased in the follow days. The SOD and POD might in a complex compensation mechanism. Both PRO and MDA content showed a notably decreasing trend in the first days and then increasing. Except for soluble sugar, the maximal and minimal values of other physiological indexesappeared on the 10 th day, but the start time of each index was different. PRO and MDA started at 0-5 d, SOD, POD and soluble sugar started at 5-10 d. Soluble protein is initiated at 15-20 d. At last, the expression of stress-related genes after waterlogging stress was detected by real-time PCR. It was preliminarily believed that the three stress-related genes, MhWRKY1, MhWRKY40b and MhMYB121 , may be involved in the response of M. hupehensis to waterlogging stress.
