摘 要:PYR1-LIKE (PYL)作为脱落酸受体,参与植物的多种逆境胁迫。从矮牵牛W115系( Petunia×hybrida ‘Mitchell’)中克隆PYL4的同源基因PhPYL4。该基因开放阅读框(Open Reading Frame,ORF)大小为645 bp,编码214个氨基酸。PhPYL4蛋白分子式为C 1022 H 1662 N 306 O 316 S 10 。该蛋白含有一个保守的疏水性配体结合结构域,属于SRPBCC超家族成员。表达分析显示:PhPYL4基因在叶片中表达量最高,根中表达量最低;盐胁迫12 h时,PhPYL4的表达量上调至对照的6倍,24 h后表达量明显下降;体积分数10% PEG处理12 h时,PhPYL4的表达量上调至对照的13倍,24 h后表达量开始下降;ABA处理12 h 时,PhPYL4的表达水平上调至对照的29倍,24 h后表达水平显著下降。研究结果揭示: PhPYL4在矮牵牛对盐、干旱及ABA等非生物胁迫的响应中具有重要作用。该研究可为进一步揭示PhPYL4在矮牵牛抗逆中的调控机制奠定理论基础。As the abscisic acid receptor,PYR1-LIKE (PYL) is involved in multiple adversity stresses.PhPYL4,the homologous gene of PYL4,was cloned from Petunia×hybrida ‘Mitchell’ in this study.PhPYL4 had an open reading frame (ORF) of 645 bp and encoded a protein of 214 amino acids.The molecular formula ofPhPYL4 protein was C 1022 H 1662 N 306 O 316 S 10 .The protein contained a conserved hydrophobic ligand binding domain and belonged to the SRPBCC superfamily.Expression analysis showed thatPhPYL4 had the highest expression level in leaves and the lowest one in roots.The expression level ofPhPYL4 increased 6 times of that of the control after 12 h under treatment of salt stress,and decreased significantly after 24 h.The expression level ofPhPYL4 increased 13 times of the control after 12 h under treatment with φ=10% PEG,and decreased at 24 h.The expression level ofPhPYL4 was up regulated 29 times of the control after ABA treatment for 12 h,and significantly decreased at 24 h.The results showed thatPhPYL4 played an important role in the response of petunia to abiotic stresses,such as salt,drought and ABA.This study lays a theoretical foundation for further revealing the regulation mechanism ofPhPYL4 in stress resistance of Petunia.
