金匮肾气丸对雄性失眠大鼠多巴胺和褪黑素的影响

摘　　要：目的 研究金匮肾气丸对雄性失眠大鼠多巴胺(DA)和褪黑素(MT)的影响。方法 60只SD雄性大鼠,随机留取10只为空白组,其余大鼠注射对氯苯丙氨酸(PCPA)造成失眠模型,随机分为模型组、唑吡坦Ⅰ组、唑吡坦Ⅱ组、肾气丸Ⅰ组、肾气丸Ⅱ组,给药14天以及停药7天后检测大鼠血中DA和MT含量。结果 给药后,与模型组比较,肾气丸组的大鼠DA含量显著降低(P<0.05),MT含量显著升高(P<0.05);停药7天后肾气丸组的大鼠DA含量较唑吡坦组显著降低(P<0.05),MT含量显著升高(P<0.05)。结论 金匮肾气丸对改善雄性失眠大鼠的睡眠有显著效果,且作用时间长;其作用机制可能与降低多巴胺、增加褪黑素含量有关。OBJECTIVE To research the Dopamine (DA) and Melatonin(MT) effects of JinGui ShenQi Pill in male insomnia rats. METHODS 60 SD male rats,randomly select 10 blank groups,and the rest of rats were injected with chlorophenylalanine (PCPA) to create insomnia model,then randomly divided into model group,zolpidem group Ⅰ,zolpidem Ⅱ group,Shenqi pill group Ⅰ,Shenqi pill Ⅱ group,detected DA and MT of rats after 14 days treatment and 7 days after drug withdrawal. RESULTS After treatment,compared with the model group,the DA content in the Shenqi Pill group was significantly lower(P <0.05),MT content increased significantly ( P<0.05);7 days after stopping the drug,compared with the zolpidem group,the DA content in the Shenqi Pill group was significantly lower( P <0.05),and the MT content was significantly increased( P <0.05). CONCLUSION JinGui Shenqi Pill can improving sleep in male insomnia rats and had long acting time;Its mechanism may be related to reducing DA and increasing MT content.
