摘  要:目的:探讨蛴螬提取物对兔光损伤视网膜变性Fas、Fasl表达的影响.方法:将30只实验性兔随机分成5组,分别为空白组、模型组、蛴螬低剂量组、蛴螬中剂量组、蛴螬高剂量组,每组6只动物,12只眼.除空白组外,其余4组均建立光损伤视网膜变性模型,在给药14d处死全部兔子后,取材做Fas、FasL免疫组化检测,采用计算机系统图像分析结果并进行统计分析,Western Blot法检测各组视网膜组织中Fas、FasL蛋白表达.结果:与模型对照组相比,蛴螬中、高剂量治疗组的Fas、FasL积分光密度与蛋白表达均降低,差异有统计学意义(P<0.05).结论:蛴螬通过降低凋亡因子Fas、FasL的表达,抑制细胞凋亡受体信号通路来发挥保护视网膜的作用,中、高剂量组效果较好.Objective:To investigate the effects of alfalfa extract on the expression of Fas and Fasl in rabbits with photodamaged retinal degeneration.Methods:Thirty experimental rabbits were randomLy divided into 5 groups:blank group,model group,low dose group,middle dose group,and high dose group,each group consisting of 6 animals and 12 eyes.Except for the blank group,the other four groups established a model of light loss retinal degeneration.After all the rabbits were sacrificed for 14 days,the Fas and FasL immunohistochemistry were performed,and the computer system image analysis resuLts were performed and statistical analysis was performed.Western Blot assay for Fas and FasL protein expression in retinal tissues of each group.Results:Compared with the model control group,the Fas and FasL integral optical density and protein expression of the middle and high dose treatment groups were decreased,and the difference was statistically significant (P<0.05).Conclusion:Grubprotects the retina by decreasing the expression of Fas and FasL and inhibiting the apoptotic death receptor signaling pathway and the medium and high dose groups performed better.
