摘　　要：目的:建立一种操作简便、准确度高、重现性好的黄芪饮片中黄芪甲苷含量检测的分析方法。方法:比较了几种不同的前处理提取方法,最终确定以回流提取和超声提取相结合,正丁醇和氨试液萃取提纯的前处理方法,结合HPLC-ELSD法进行黄芪甲苷的含量测定。结果:黄芪甲苷在0.06~1.9 mg/mL浓度范围内线性关系良好(r=0.999 8),平均回收率为100.16%,RSD=2.63%(n=5)。结论:改进后的方法准确、高效、重现性良好,适用于黄芪饮片中黄芪甲苷的含量检测。Objective:To establish a simple,accurate,and reproducible quantitative method for determination of Astragaloside Ⅳ in Astragali Radix.Methods:Several different extraction methods were used and compared.Astragaloside Ⅳ was extracted by the method of hot reflux extraction and ultrasonic extraction,and the extracting solution was purified by n-butanol and ammonia solution.The content of Astragaloside Ⅳ was determined by HPLC-ELSD.Results:The assay was validated in the concentration range of 0.06-1.92 mg/mL(r=0.999 8).The average recovery was 100.16% with RSD=2.63%(n=5).Conclusion:The improved method is accurate,effective> reproducible,and can be used for determination of Astragaloside Ⅳ in Astragali Radix.
