摘  要:黑木耳(Auric ularia auric ula-judae)是一类栽培广泛的食药兼用菌,为调查黑龙江省东部地区市售黑木耳的种质资源,收集了东宁县黑木耳批发市场黑木耳干品11个和牡丹江市售黑木耳试管菌种6株.采用组织分离方法获得菌丝体,提取菌丝体DNA,扩增ITS片段并测序.测序结果发现,经比对,17株供试黑木耳菌株ITS序列同源性均在99%~100%,其中16株的ITS序列长度为559 bp;系统发育分析表明,供试17种黑木耳菌株可分为7种不同类型,表明市售黑木耳菌种存在商品名不同但实际为同一品种的情况.以上结果为黑木耳种质资源调查及当地黑木耳品种引进提供理论指导.A uricularia auricula-judae is a kind of widely cultivated edible and medicinal fungi. To investigate the germplasm resources of A. auricula-judae in the east area of Heilongjiang province, 11 dried A. auricula-judae samples from Dongning wholesale market and 6 tubes of A. auricula-judae strain from Mudanjiang were collected. The mycelium were obtained by sep aration method, mycelium DNA were extracted, ITS sequences were obtained and then sequenced. The results showed that, the similarities of the 17 ITS sequences were 99%-100% through comparison. Among them, the length of 16 ITS sequences were 559 bp. Phylogenetic analysis showed that, 17 tested A. auricula-judae strains could devide into 7 different types, which means some of the A. auricula-judae strains have different trade name but indeed, they are the same cultivar. The results provide the oretical direction for investigating A. auricula-judae germplasm resources and local introduction.
