摘  要:目的:研究红景天苷在大鼠肝微粒体中的酶促反应动力学。方法:采用UPLC-MS测定肝微粒体孵育体系中红景天苷的含量,色谱柱为WatersBEHC18(2.1mm×50mm,1.7μm)柱,流动相为乙腈-0.1%甲酸水溶液,梯度洗脱,采用电喷雾电离源,扫描方式为选择离子监测模式。根据Lineweaver-Burk方程分析数据求算酶促动力学参数。结果:红景天苷在线性范围内线性关系良好(r=0.999,9),准确度、精密度、回收率、稳定性和基质效应均符合要求。红景天苷的酶促动力学参数Vmax=58μmol/(mg·h),Km=41.07μmol/mL,固有清除率(CLint)为0.708mL/(mg·h)。结论:建立的分析方法快速、准确、灵敏,可满足红景天苷在大鼠肝微粒体中的酶促动力研究,为进一步研究红景天苷代谢性相互作用奠定了基础。Objective:To study the enzymatic kinetics of Salidroside in rat liver microsomes.Methods:Salidroside was determined by UPLC-MS with WATERS ACQUITY UPLC BEH C18 column.The mobile phase, consisting of acetonitrile and 0.1% aqueous formic acid,was programmed as a linear gradient.The quantitation was done by selective ion monitoring mode of electrospray ionization.The enzymes kinetics value was evaluated by graphical analysis with Lineweaver-Burk double reciprocal plots.Results:The relationship between the concentration and the peak areas of salidroside was linear (r=0.999,9).The precisions,accuracy,extract recoveries, matrix effect and stability meet the requirements.The corresponding enzymatic kinetic parameters as follows:Vmax=58 μmol/(mg·h),Km=41.07 μmol/mL,CLint=0.708 mL/(mg·h).Conclusion:The method was shown to be rapid,sensitive and suitable for study the enzyme kinetics of salidroside in rat liver microsomes.Enzyme kinetic parameters obtained of salidroside provide important parameters for the further study.
