摘 要:溶菌酶是具有抗菌、消炎作用的生物活性物质,应用极其广泛。以市售溶菌酶为原料,采用PEG2000/K2HPO4双水相体系萃取溶菌酶,考查了不同K2HPO4浓度、不同PEG2000浓度、不同溶菌酶浓度和不同用量NaCl4个因素对溶菌酶在双水相体系中的分配情况。结果表明,萃取溶菌酶的最佳工艺条件为在室温条件下,当质量分数为45%(W/W)磷酸氢二钾3mL,质量分数为45%(W/W)PEG20005mL,质量浓度为1mg/mL溶菌酶2mL,NaCl用量为0.2g/10mL时,溶菌酶主要分配在双水相体系的上相,相比为1,分配系数为6.53,萃取率为75.3%。Lysozyme is an anti-bacterial and anti-inflammatory,biologically active substances,and was used extremely widely.In this paper,using commercially available lysozyme as raw material,and using PEG 2000/K2HPO4 aqueous two-phase system to extract lysozyme.Four different factors(different concentrations of K2HPO4,different concentrations of PEG 2000,different concentrations of lysozyme and different mass of NaCl)were investigated on the influence of distribution of lysozyme in aqueous two-phase system.The optimum conditions of extracting lysozyme:at room temperature,when 3 mL 45%(W/W)K2HPO4,5 mL 45%(W/W)PEG 2000,2 mL 1 mg/mL lysozyme,and 0.2 g/10 mL NaCl,Lysozyme are mainly in the upper-phase of aqueous two-phase system,proportion was 1,the distribution coefficient was 6.53,the extraction rate was 75.3%.
