摘 要:为了探究2019年1月份海南省乐东县某养殖场所培育的珍珠龙胆石斑鱼鱼苗发生大规模死亡的病因,本研究对发病情况进行了调查、临床诊断和实验室诊断.研究结果显示:该病并未在主要发病季节发生,但出现了典型的病毒性神经坏死病(viral nervous necrosis,VNN)症状,其症状为食欲降低、行为异常、转圈、鱼体发黑、体况差等,累计死亡率达62%.本研究针对患病鱼体进行了寄生虫学检查,结果为阴性;组织病理学结果显示:患病鱼的视网膜和脑组织空泡化严重;应用世界动物卫生组织(OIE)推荐的VNN特异性检测引物,运用RT-PCR检测方法,扩增出病毒性神经坏死病毒(nervous necrosis virus,NNV)特异性的421bp基因片段;运用虹彩病毒科蛙病毒(Ranavirus)、细胞肿大病毒(Megalocytivirus)检测引物,采用PCR方法排除了混合感染,并初步诊断该病为病毒性神经坏死病,此外,针对石斑鱼养殖后期的病害防御提出了合理的措施。In the report,in order to investigate the causes of mass death of the Hybrid grouper fry ( Epinephelus fuscoguttatus♀× E.lanceolatus♂),which was cultured in a farm of Ledong County,Hainan Province in January 2019,the morbidity,clinical diagnosis and laboratory diagnosis were carried out.The results showed that the disease of Hybrid grouper fry did not erupt in the main onset season,however,the typical symptoms of viral nervous necrosis (VNN) occurred.Its symptoms were loss of appetite,abnormal behavior,circle turning,blackening body,poor health and subsequent death with a cumulative mortality rate of 62%.The parasitological examination result of the diseased fish was negative.The primers recommended by the World Animal Health Organization (OIE) were used for the detection of NNV,a 421 bp specific gene fragment of nervous necrosis virus (NNV) was amplified by RT-PCR.The specific primers of Rana virus and Megalocytivirus were used to exclude the mixed infection.These results indicated that the disease was VNN infection.After diagnosis,the reasonable measures were put forward for the further prevention.
