摘  要:为研究树莓的工厂化育苗技术,提高树莓育苗质量和效率,以树莓品种‘海尔特兹’带腋芽的茎段为试验材料,通过对其腋芽初代培养、茎段增殖、叶柄不定芽和根的诱导等过程中植物生长物质合理组合,及防止茎段褐化的最适pH值进行筛选,建立‘海尔特兹’树莓组织培养及其叶柄再生体系。结果表明,腋芽诱导的最佳培养基为MS+1.0mg·L^-16-BA+1.0mg·L^-1IAA,萌芽率100%;茎段最佳增殖培养基为MS+0.5mg·L^-16-BA+0.1mg·L^-1NAA,pH值为5.70,增殖系数为5.0;诱导叶柄产生愈伤组织再生不定芽的最佳培养基为MS+2.0mg·L^-1TDZ+1.0mg·L^-1IBA,其愈伤组织诱导率为100%,不定芽再生率为53%;幼苗最佳生根培养基为1/2MS+0.7mg·L^-1IBA,生根率达95%。In order to improve the quality and efficiency of industrial seedling cultivation of raspberry, stem segments with axillary buds of the raspberry cultivar ’Heritage’ were used as experiment material, the regeneration system of leaf petioles was established through screening of the reasonable plant growth substances combination in the process of axillary bud primary culture, stem segment proliferation, petiole adventitious bud and root induction, and the optimum pH value to prevent stem segment browning. The results showed that the optimum medium for axillary bud induction was MS+1.0 mg·L-16-BA+1.0 mg·L-1 IAA, and the germination rate was 100%;the optimum medium for stem segment proliferation was MS+0.5 mg·L-16-BA+0.1 mg·L-1 NAA, and the pH value was 5.70, which the multiplication coefficient was 5.0;the optimum medium for petiole callus regeneration was MS+2.0 mg·L-1 TDZ+1.0 mg·L-1 IBA,which the induction rate of callus was 100%, and the regeneration rate of adventitious bud was 53%;the optimum medium for rooting was 1/2 MS+0.7 mg·L-1 IBA, and the rooting rate was 95%.
